| Ginkgo biloba L. is the only extant plant of Ginkgoales, and has high economic and scientific value.In this study, the difference in genetic among 40 Ginkgo male clones which grown at 11 provinces were analyzed by the RAPD markers. Variation of seeds characters, seedling physiology indexes, seedling growth characters and medicine quality indexes among controlled-pollination hybrid progeny were primary studied. The main results are as following:1. Twelve pairs of primers with high stability and good repeat ability were selected from 100 or more pairs of primers. Among 96 fragments generated by 12 selected primers, 41 appeared to be polymorphic, the polymorphic locus percentage were 42%, average effective number of alleles, average Nei's gene diversity, average Shannon's information index were 1.6440, 0.3752 and 0.5562, respectively. According to the cluster analysis, the relative relationship among different clones was correlated with geographical distribution.2. There were significant differences in the seed characters among hybrid progeny. The variance of weight traits was great; while the variance of shape traits was low. The relative analysis of seeds characters showed that selecting great weight per seed, nut width, nut thickness, seed width, seed length was favorable to selecting cultivars of great weigh per nut.3. The difference in the physiology indexes among hybrid progeny were significant, theindexes changed monthly. The correlation analysis showed that net photosynthetic rate, transpiration rate, NR activity, and PPO activity could be selected as selecting indexes for early-selection.4. There were significant differences in growth characters among hybrid progeny. Hybridprogeny were good at seedling height, seedling biomass and so on main growth characters in comparison with contrast.5. Ten indexes were selected as early selecting indexes by correlation analysis. With the help of factor analysis and cluster analysis, five superior families in growth characters wereinitially selected.6. The study showed that there were significant differences in the medicine quality indexes among hybrid progeny. For total flavonoid contents, quercetin contents>kaemplerol contents> isorhamnetin contents; For total terpene lactone contents,ginkgolide A contents >bilobalide contents>ginkgolide B contents>ginkgolide C contents. Variation analysis showed that selecting medicine quality indexes of leaves and leaves biomass at the same time would gain better effect. Eight superior families in leaves were selected by cluster analysis. There were significant differences in the 6 microelements in leaves. |
