Cyc and Dich, belong to the Cyc/Tb1 sub-family of TCP-domain genes, were responsible for the control of floral asymmetry in the snapdragon (Antirrhinum majus). Several homologies of Cyc and Dich in Soybean (Glycine max cv. Jilin21) and Lotus joponicus (Gifu) were identified. In situ hybridization show that: when the floral primordium initiated from the inflorescence primordium, Socyc1A and Socyc2 express transiently in the dorsal region of floral primordium, either in axillary inflorescence or in terminal inflorescence of soybean. After the floral primordium separated completely from the inflorescece meristem, Socyc1B expressed in the dorsal region of floral bud, at last, mainly in the dorsal petal and the dorsal side of the base of floral bud. During the floral development of soybean, Socyc1A and Socyc2 both involved in the construction of the dorsal-ventral axis in the early stage. However, in the later stage, Socyc1B express maintains the axis. Soybean shares the same genetic mechanism in controlling the asymmetric development of flower with snapdragon. But in soybean, the function of TCP domain genes still has something different with the function of Cyc and Dich in snapdragon. In the terminal inflorescence, Socyc2 expressed not only in the dorsal region of the floral primordium, but also in the junction side in the terminal inflorescence meristem. Maybe the function of Socyc2 was involved with the initiation of the floral primordium, and the structure of the inflorescence.Ljcyc3, Ljcyc5 genes belong to Cyc/Tb1 sub-family of TCP domain genes. Ljcyc5 expressed in the base of second inflorescence, which is different with any other TCP domain genes. 86 T0 plants of Ljcyc5 antisense from 27 independent callus, 56 T0 plants of Ljcyc3 antisense from 18 independent callus have been got. The detection of GUS shows that we have got the transgenic plants with T-DNA insertion.In order to research the molecular mechanism of flower development in Legume, we construct the mutant library of Lotus joponicus. Many M2 mutants were identified. Three lines of keeled wing mutants were identified from the mutant library. The shape and the epidermis cell configuration of the wings of kw1 and kw3 exactly resemble that of the keels. kw1 is a mutant of single gene and is alleic with kw2. The F1 of kw1 and Ljcyc2 antisense transgenic plant have the phenotype of keeled wings, show that the Kw gene has interaction with TCP-domain genes.
