In order to investigate the immunomodulation and anti-oxidation of potentilla anserine polysaccharide (PAP), the effect of PAP on immunological function, activities of oxygen radical genesis enzyme in tissue and organ, the action of cleaning oxygen radical, and the action of resisting apoptosis induced by hydrogen peroxide (H2O2) in vivo and in vitro were studied by the techniques of enzyme-linked immunosorbent assay (ELISA), microculture tetrazolium assay (MTT), fluorescence spectrophotometry, radioimmunoassay, phase contrast microscope, laser scanning confocal microscope (LSCM), DNA agarose gel electrophoresis and flow cytometry assays etc. The results were as follows: (1) PAP promoted proliferation of splenocyte induced by lipopolysaccharide (LPS) or concanavalin A (ConA) with the prolongation of culture time and the increasement of PAP concentration, and the splenocyte proliferation were dose-dependant and time-dependant. PAP also promoted the index of spleen inhibited by cyclophosphamide (Cy), but had no effect on that of thymus. Moreover, PAP promoted secretion of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), interferon-y (IFN-γ) in immuno-organ. (2) The concentration of 200 μg/mL PAP could increase the content of reduced glutathione (GSH), which was a signal substance of splenocyte, but had no significant effect on the content of GSSG, and modulated the oxidation-reduction to reduction state according to the value of GSH/GSSG. (3) With the culture time prolonging and concentration of PAP increasing, the content of signal molecular nitric oxide (NO) produced in splenocyte were increased, and then NO promoted the synthesis of cGMP through activating guanylate cyclase (Gc).
