Mandarin fish siniperca chuatsi is an important economic fish widely cultured in China. Because it tastes delicious and high protein-containing, so it is favored by most of Chinese customers. However, serious viral diseases have become one of limiting factors for the aquaculture industry of mandarin fish. Based on the observation of siniperca chuatsi spherical virus (SCSV), siniperca chuatsi rhabdovirus (SCRV), siniperca chuatsi baculovirus (SCBV) from diseased mandarin fish tissues and the confirmation of ultrastructural changes on host cells caused by coinfection of SCSV and SCRV, virus was separated by plaque-isolation associated with electron microscopic technology in order to understand the essence of the pathogen and prevention and cure the disease. Studies on characterization of SCRV, sequencing of the complete genome, analysis and expression of the functional genes were carried out. The main research methods and results were described as follows:1. Plaque-isolation of SCRV combined with electron microscopic technology.Mandarin fish tissues with typical haemorrhagic syndrome of viral infection were prepared into tissue suspensions which were serial 10-fold diluted from 10-1 to 10-4 and inoculated onto grass carp fins (GCF) cells respectively. Cells at 12, 18, 24, 30 and 36 h postinfection were collected and prepared into ultrathin sections followed by electron microscopy observation.
