In this study,the MSTN gene sequence(GeneBank Accession: DQ167575) was amplified and sequenced by PCR among different breeds including main local goat in China and introduced breeds. The composition of nucleotide and encoded amino acid were analyzed. The molecular evolution trees were established by Bioedit,Phylip etc.. The PCR-RFLP analysis of the part sequence of MSTN gene in different goat breeds were studied.The results were as follows:1. The MSTN gene sequence was amplified by PCR in goat.The DNA sequence was 5211bp in length,including three extrons and two introns. The length of three extrons was 373bp, 374bp and 381bp respectively. The length of two introns was 1834bp and 2027bp respectively.2. Prediction of the binding site of transfactor found abundant heat shock response elements in the squence.The nucleotide identity of goat MSTN genes with cattle, pig, horse, human, baboon, dog, red fox, mouse, turkey, partridge, duck, goose, chicken, pigeon, atlantic salmon, weever, croceine croaker, zebrafish, channel catfish was 96.37, 94.68, 93.53, 92.38, 92.29, 91.22, 90.60, 89.36, 82.98, 82.98, 82.89, 82.80, 82.35, 81.91, 38.21, 29.70, 27.59, 24.02, 23.59 percent respectively. There were 375 amino acid sequence in goat MSTN protein. The mature peptide coded by goat MSTN gene shared the unique feature with that of another mammal, as cleavage at the predicted RSRR proteolytic processing site producing 127 amino acid peptide which contained 9 cysteine residues. All this results indicated the relatively high conservation of MSTN gene among different species.3. PCR-RFLP analysis was conducted on the partial nucleotide sequence of goat MSTN gene. The result showed that the sequence of the extron 2 and extron 3 were identical in goat breeds.But eight polymorphic sites were found in the 5′UTR, extron 1 and intron 2 region. One TTTTA insert at nucleotide position 217,one C→T transversion at nucleotide position 3617, one C→A transversion at nucleotide position 3726, one G→A transversion at nucleotide position 4794, one GCGAT→AGAAG transversion at nucleotide position 4600-4604, two T→C transversion at nucleotide position 3783 and 4280, While the T→A transition at position 345 result in no changes of the amino acid substitution.
