In the seed orchard of Pinus tabulaeformis Carr. in Xingcheng, Liaoning Province, some researchers had found that the clone 28 mutated-its female cone grew normally, while the ovules aborted because that the female gametophyte couldn't form due to the mitosis failure of free nuclei in female gametophyte. The clone 28 is an excellent material for ovule development mechanism research of woody plants with its steady genetic characteristic of female sterility. In this paper, we studied the hormone level and the proteome of female-sterile clone and hoped to discover the reason of ovule abortion.Hormones are important factors in regulating plant growth and development. Content changes of several endogenous plant hormones in female-sterile clone and normal clone of Pinus Tabulaeformis Carr. were analyzed by enzyme-linked immunosorbant assay during the key period of ovules abortion in mutant clone. The results indicated that the high level of IAA was important for free nuclei entering into the cell cycle again after dormancy. The high level of ZR and low level of IAA were crucial for normal mitosis of free nuclei. The high level of ABA and deficit of IAA and ZR may be the reasons of ovule abortion in female-sterile clone of Pinus Tabulaeformis Carr. because free nuclei could not enter into the cell cycle normally.Proteins are expression products of functional genes. They affect growth and development of plant directly since they are executants of biological function. In order to assure the reproducibility of results, establishing a steady method is very crucial in proteomic study. In this study, we optimized the extraction method of ovule proteins, the conditions of 2-dimensional electrophoresis and mass analysis of peptide mass fingerprinting. The proteomic technique of ovule proteins from Pinus tabulaeformis Carr. was established..On the basic of establishment of proteomic technique, the proteins of ovule from female-sterile clone and normal clone were separated with 2-dimensional electrophoresis (2-DE) during the key period of ovules abortion in mutant clone. There were 20 proteins expressed only in normal clone and 13 proteins expressed only in female-sterile clone. The volume of 25 proteins was different between normal clone and female-sterile clone. The molecular weight of these proteins distributed in 25kDa-35kDa and 60kDa-70kDa mainly and pI distributed in 5.4-6.0 and 6.8-7.5.
