Open-field behavior test and temperament test were carried out on 204 two-month old puppies of German Shepherd dog, Labrador Retrieve and English Springer Spaniel. Meanwhile, genes of MAO-A, MAO-B, COMT, TH, DpH, DR2 and 5-HTT were chosen as the candidate genes that affect canine behavior traits. Single nucleotide polymorphisms and microsatellite markers in candidate genes were detected and the genetic effect of molecular markers of the candidate genes on behavior traits were determined. The purpose of the current study is to study the methods of dog behavior tests, compare the differences of behavior traits among dog breeds, find the molecular markers related to canine behavior traits and provide methods for molecular marker assistant breeding. The main contents presented as follows.1 Comparative studies of dog behavior tests in German Shepherd dog, Labrador Retrieve and English Springer SpanielGerman Shepherd dog, Labrador Retrieve and English Springer Spaniel were often used as working dogs, although their temperament and behavior were different. German Shepherd dog was stable, fearless and alert, obedient and eager to learn. These dogs were all-powerful working dogs and could be used as criminal recon, maintaining public security, searching drugs and exploder. Labrador Retrieve was gentle and friendly, patient and willing to fetch. These animals were excellent guide dogs and could be used as search dog and succor dog. While, English Springer Spaniel was active and good at searching. The experimental dogs included 42 German Shepherd dogs, 71 Labrador Retrieves and 91 English Springer Spaniels in the present study. Behavior traits in three breeds, such as excitability, courage and desire to fetch, were evaluated with open-field behavior test and . temperament test. The differences between male and female were compared. The effect of age and parity of bitch on behavior variables were analyzed. Statistics analyses were conducted by using SAS 8.2.â‘  Open-field test German Shepherd dog, Labrador Retrieve and English Springer Spaniel showed significantly different behaviors in open-field test.
