Detection Procedure of Chicken Spleen Transfer Factor(TF) including components detection and physical &chemistry qualities was established and Evaluated for 9 batchs of TF. Three components of TF were detected by HCLP, As standard, insulin is used to ensure whether some substances over 5000 exist in TF. Peak area lower insulin saving time should be lower1%.Different batchs and original TF have the same peak uptrend on Hydrolysis amino acid in TF. But different in Dissositive Amino acid. Dissositive Amino acid could be used as one of standard of quality control. The sum of dissositive Amino acid should be higher 2mg.Separating TF by Sephadex G-25 and collecting the three different components, these three components were determined by MTT colorimetric method. The result indicated that the mixture distilled from chicken spleen has three components. All the three ingredients and crude TF can improve the transformation of immunosuppressant murine T lymphocytes, which is different from normal murine T lymphocytes that the first ingredient and crude can improve it and the second ingredient has no result but the third one could decrease the transformation T lymphocytes. which is different from the report of TF applied in man.Ultra filter and column chromatography combination technology was used to extract TF. Compared with the traditional technology, Ultra filter obtains higher polypeptide and ribose, which can be used in industrilizational production. Compared with the changes of polypeptide and ribose of chicken spleen TF stored at 4℃and -20℃, 4℃is better than -20℃. Polypeptide and ribose can decrease after keeping in a long time. The best store time should be lower 12 months.Compared with different using does and using instructions between TF and vaccine plus TF, the result is different in Average Daily Gain(ADG), mortality, rate of lymphocytes, immuned organ index and phagocytosis index of chicken macrophage. The result indicates that TF has no clear function in healthy broilers, but in unhealthy broilers, TF could improve immunity, ADG and insistence of disease. Compared with vaccine group and vaccine plus TF group, the difference is not clear, Compared with mortality, ADG and phagocytosis index of broiler macrophage between TF and control, TF group is obviously better than control, the same like vaccine group. TF can be associated with vaccine. It is suggested that TF could improve performance and 0.5ml per chicken in oral is best.Clinical experiments shows that TF had an obvious effect in improving performance and preventing disease in unhealthy poultry. TF has been succeded in broilers,layers and ducks.According to physical & chemistry qualities of TF and immune index, clinical index, Detection Procedure of Chicken Spleen Transfer Factor(TF) was formulated and used in guiding production, which can provide basis of TF.
