Studies on the structures of PIP1 and PIP2 indicated that:(1) PIP1 is composed of the monosaccharide of glucose, galactose and mannose,and the molar ratio is 3.70 : 4.06 : 1.00. The glucosidic bonds of PIP1 wereβconfiguration . It had few branch. The main chain is made up of a great mass of glucose(1→3)and few mannose(1→4); The side chain is composed of glucose(1→3)and galactose(1→6); The non-reduced end is composed of galactose and glucose . The branch spot is 6-O of glucose (1→3,6 ) and Mannose(1→4,6). On average, there are 3 branches among 20 residues.(2) PIP1 is mainly composed of glucose, and few of galactose and mannose.Its glucosidic bonds wereβconfiguration. It had few branch. On average, there were 1.5 branches every 10 hexose residues. PIP2 possessed a backbone of (1→4)-linked glucose residues, with substitution at O-3 by (1→3)-linked glucose. The side chains was composed of (1→3)-linked glucose with different degree of polymerization. The end was composed of Glc.PIP1-S and PIP2-S(Sulfated PIP1 and PIP2) were obtained with method of chlorosulfonic acid-pyridine. The SO4-2 contents of PIP1-S and PIP2-S is 48.00% and 52.75% respectively.The antitumor activity of PIP and the sulfated derivatives were studied in vitro. The results have shown that both PIP and sulfated derivatives had no antitumor activity.The transformation of lymphocytes activities of PIP and the sulfated derivatives were examined. The results indicated that the PIP and the sulfated derivatives exhibited high activity. The activity of the sulfated derivatives were more than that of the PIP.Based on the findings of this study,the results suggest that immunomodulation was the mechanism of the antitumor activity of PIP. So it was believed that the existence of 1,3-linked-Glc in the main and side chains is the main reason for the increased antitumor activity.
