To develop high frequency regeneration system of soybean tissue culture, weperformed tissue culture of soybean cotyledonary node to find out some factors thatinfluence differentiation of soybean cotyledonary node to adventitious buds andseveral different soybean cultivars and lines were used to test genotype effect onregeneration frequency.The conclusions are as the followings:1, Low concentration of BA favours the differentiation of soybean cotyledonarynode.2, Glutamine (added to media before autoclave) can prompt differentiation ofsoybean cotyledonary node.3, AgNO3 enhances effect on regeneration of soybean cotyledonary node.4, Planting cotyledonary node with 1cm length of hypocotyl (the hypocotyl is put inmedia) can induce soybean cotyledonary node to form adventitious buds directly.5, Differences in regeneration capacity result from different genotypes.6, The medium cotaining B5+1.5mg/L BA+50mg/L Glu+50mg/L Asp+6mg/LAgNO3+3% sucrose is good medium for the differentiation of soybean cotyledonarynode.
