Gestational Diabetes Mellitus (GDM) is the most common metabolic disturbant complications in pregnancy duration, and do prodigious damage to the pregnant women and foetus. So early detection and early treatment the disease could effectly reduce the complication, improve the pregnancy outcome. But the methods of early screening reliably and conveniently are absent at present. Proteomics is a new way to study the proteins, it can screening candidate protein quickly, extensively and comprehensively. And it had already become a new pathway to find biological markers. However it was rarely used in clinical because of complexly operating and highly costing. Urinary proteome was not studied in patients of GDM. Therefore the analysis of urinary proteins is meaningful for biomarker discovery, early screening method establishing and physiological investigation.Human urine, a readily available atraumatic body fluid, contains many proteins and peptides that perform biological functions, and hence, can be potentially useful in the screening and diagnosis of many diseases. Recent studies have already revealed several urinay proteins related to diseases, including type2diabetes mellitus (T2DM).The aim of our study was to establish urinary proteomic spectrum in healthy pregnant women and GDM women of25-30gestational weeks. And to find out differential proteins by contrasting the groups. We precipitated proteins by ethanol and measured the concentration by Bradford method. Both group included9pregnant women. The urinary samples of both group were mixed respectively. After microwave-assisted digestion by trypsin, we used liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) to identify the proteins.We identified837urinary proteins in healthy pregnant women,791in GDM patients and983proteins in total. We found14differential proteins between healthy and GDM groups. Compare to the healthy group, GRN、FGB、CPVL、S100A11etc. were found upregulated in GDM groups, and HSPA8、LTF、CA1were found downregulated. Many of these proteins were involved in gluconic metabolism. Next step, we should Validate the proteins and expand the sample size for clinical guidance.
