There has been many researches focusing on absorption and digestion of thegastrointestinal tract, with the deepening of the basic and clinical study,gastrointestinal mucosa barrier function becomes more and more important. Thestability of intestinal mucosa barrier function directly affect the occurrence,development and outcome of digestive diseases. And play an important role ofintestinal mucosa barrier in resisting invasion of pathogenic factors outsideof the body. Intestinal mucosa barrier consists of epithelial junctions, mucussecretion, resident microflora and associated lymphoid tissue etc.The mucin (MUC)is a kind of glycoprotein. MUC2is also called intestinal mucin, as being secretedby the intestinal goblet cells. It is the main part of the normal intestinalmucus, and with high viscosity under certain conditions. MUC3is a kind oftransmembrane mucin, it is mainly expressed in the intestine and colonic villousepithelial cells. MUC5AC is mainly expressed in gastric mucosa, and in certainpathological conditions such as inflammatory bowel disease, cancer, it isexpressed in intestinal mucosa. Mucin can lubricate the intestines, form theprotective barrier in intestinal mucosa, and also has involve in the cellularsignal transmission and antigen presenting.Intestinal trefoil factor (TFF3)exists widely in the intestine, and form the net structure with mucin to protectthe mucosa. TFF3also can drive cell migration to repair the intestinal mucosa.And mucin and trefoil factor have played a positive role in intestinal barrier.The resident flora form the biological barrier, which can prevent pathogen andkeep the balance of micro environment. T lymphocytes and sIgA can regulate theimmune function and improve the Intestinal mucosa barrier through suppressingthe immune response, eliminating pathogenic factor, and neutralizing endotoxin.These kinds of barriers are mutual coordinated and interdependent with others.They form a complete barrier system to against various pathogenic factors.According to The theory of traditional Chinese medicine, spleen is acquiredsource, it has important physiological functions, and can provide the materialfoundation for other organs, and resist the evil. The function of spleen directlyaffects the ability of body defense capability, therefore it has the theory of“the spleen acting as the protector of the body ".As gastrointestinal defense mechanism, intestinal mucosa barrier has thesame physiological and pathological basis with traditional Chinese medicinetheory “the spleen acting as the protector of the body ". This research is basedon this theoretical relation, observes the changes of intestinal mucosa barrierfunction of rats with spleen asthenia syndrome from different angles, andevaluates the effects of classical decoction, Sijunzi Decoction which caninvigorate the spleen and replenish qi. Further explores the influence ofinsufficiency of the spleen syndrome on intestinal mucosal barrier function andthe prevention and treatment effects of traditional Chinese medicine.Section1: The experimental study on intestinal mucosa changesof rats with spleen asthenia syndrome and the effect of SijunziDecoctionPurpose: To observe the pathological changes of intestinal mucosa in spleenasthenia syndrome rats and the effects of Sijunzi Decoction which can invigoratethe spleen and replenish qi.Material and method:fourty-eight SD rats,2-3months of age, were dividedinto4groups randomly:the normal control group, the spleen asthenia syndromemodel group of TCM, the natural rehabilitation model group, and the SijunziDecoction control group.killed the rats, and made the section of intestinalmucosa. Results:of400X light microscopy gastrointestinal pathology morphologicalobservation showed:1. The normal control group: intestinal mucosa structure of each layer iscomplete, the texture is clear; and there is no inflammatory reaction anddamage on it.2. The spleen asthenia syndrome model group of TCM: mucosa structural disorder,visible necrosis exudation and other pathological changes; The intrinsiclayer and submucosa vascular dilatae and congest, even hemorrhage. Plasmacells are infiltrating downwards.3. The natural rehabilitation model group: Intestinal mucosal defect, visibleinflammatory cell infiltration, mucosal edema.4. the Sijunzi Decoction control group: returned to normal largely, the mucousglands were arranged in order.Conclusion:According to the etiology theory of TCM, the overstrain, improperdiet and drug induced injury combined method can successfully induced spleenasthenia syndrome model in rats. In the state of spleen asthenia syndrome, thestructural integrity of intestinal mucosa is damaged. While, Chinese decoction,which has the function of invigorate the spleen and replenish qi, SijunziDecoction can repair it.Section2: The experimental study on intestinal mucosa mucin(MUC2, MUC3, MUC5AC) and trefoil factor (TFF3) changes of ratswith spleen asthenia syndrome and the effect of SijunziDecoction Purpose: Toobservethechangesofintestinalmucosamucin(MUC2,MUC3,MUC5AC)and trefoil factor (TFF3) expression in spleen asthenia syndrome rats and theeffects of Sijunzi Decoction which can invigorate the spleen and replenish qi.From the view of mechanical, chemical barrier function of intestinal mucosa,to explore the mechanism of the traditional Chinese medicine theory " the spleenacting as the protector of the body ".Material and method:detect the Protein expression of mucin (MUC2, MUC3,MUC5AC) and trefoil factor (TFF3) in normal control group and other spleenasthenia syndrome model groups of intestinal mucosa with Western-Blot methodand immunohistochemical method, detect the expression of mRNA with RT-PCRmethod.Results:1. Compared with the normal control group, the protein expression of MUC2, MUC3in intestinal tissue of the spleen asthenia syndrome model group of TCM, thenatural rehabilitation model group,and the Sijunzi Decoction control groupwere decreased (P<0.01),MUC5AC expressed, has statistically significantdifference. With the therapeutically treatment of Sijunzi Decoction, theprotein expression of MUC2, MUC3in intestinal tissue of the Sijunzi Decoctioncontrol group increased better than the spleen asthenia syndrome model groupof TCM and the natural rehabilitation model group, MUC5AC decreased,hasstatistically significant difference,(P<0.01or P<0.05).2. The mRNA expression of MUC2in intestinal tissue of the spleen astheniasyndrome model group of TCM, the natural rehabilitation model group,and theSijunzi Decoction control group were decreased (P<0.01), MUC5AC expressed,compared with the normal control group, has statistically significantdifference. With the therapeutically treatment of Sijunzi Decoction, theprotein expression of MUC2in intestinal tissue of the Sijunzi Decoction control group increased better than the spleen asthenia syndrome model groupof TCM and the natural rehabilitation model group, MUC5AC decreased, hasstatistically significant difference,(P<0.01).3. Compared with the normal control group, the protein expression of trefoilfactor (TFF3) in intestinal tissue of the spleen asthenia syndrome model groupof TCM, the natural rehabilitation model group,and the Sijunzi Decoctioncontrol group were decreased (P<0.01), has statistically significantdifference. With the therapeutically treatment of Sijunzi Decoction, theprotein expression of trefoil factor (TFF3) in intestinal tissue of theSijunzi Decoction control group increased better than the spleen astheniasyndrome model group of TCM and the natural rehabilitation model group, hasstatistically significant difference,(P<0.01or P<0.05).4. The mRNA expression of trefoil factor (TFF3) in intestinal tissue of thespleen asthenia syndrome model group of TCM, the natural rehabilitation modelgroup,and the Sijunzi Decoction control group were decreased (P<0.01),compared with the normal control group, has statistically significantdifference. With the therapeutically treatment of Sijunzi Decoction, theprotein expression of trefoil factor (TFF3) in intestinal tissue of theSijunzi Decoction control group increased better than the spleen astheniasyndrome model group of TCM and the natural rehabilitation model group, hasstatistically significant difference,(P<0.01).Conclusion: Inthestateofspleenastheniasyndrome,secretionofpartmucinin intestinal mucosa reduce. Through the treatment of Sijunzi Decoction, thesecretion increased. That might be concluded that the Sijunzi Decoction canrepair the intestinal mucosa mechanical, chemical barrier of rats. Section3: The experimental study on serum endotoxin changes ofrats with spleen asthenia syndrome and the effect of SijunziDecoctionPurpose: Toobservethechangesofserumendotoxininspleenastheniasyndromerats and the effects of Sijunzi Decoction which can invigorate the spleen andreplenish qi. From the view of biologica barrier function of intestinal mucosa,to explore the mechanism of the traditional Chinese medicine theory " the spleenacting as the protector of the body ".Material and method:detect the serum endotoxin in normal control groupand other spleen asthenia syndrome model groups of intestinal mucosa with ELISAmethod.Results:Compared with the normal control group, serum endotoxin of the spleenasthenia syndrome model group of TCM, the natural rehabilitation model group,andthe Sijunzi Decoction control group were increased (P<0.01), hasstatistically significant difference. With the therapeutically treatment ofSijunzi Decoction, serum endotoxin of the Sijunzi Decoction control groupdecreased better than the spleen asthenia syndrome model group of TCM and thenatural rehabilitation model group, has statistically significantdifference,(P<0.01or P<0.05).Conclusion: Inthestateofspleenastheniasyndrome,the biologicabarrierfunction of intestinal mucosa is damaged, which leads to intestinal bacteriaand endotoxin translocate. Sijunzi Decoction can improve the endotoxemia, andrepair the biologica barrier function of intestinal mucosa of rats. Section4: The experimental study on intestinal mucosa secretionof sIgA and T lymphocyte (CD3)changes of rats with spleenasthenia syndrome and the effect of Sijunzi DecoctionPurpose: To observe the changes of secretion of sIgA and T lymphocyte (CD3)in spleen asthenia syndrome rats and the effects of Sijunzi Decoction which caninvigorate the spleen and replenish qi. From the view of immune barrier functionof intestinal mucosa, to explore the effect of immunity function.Material and method:Detect the secretion of sIgA in normal control groupand other spleen asthenia syndrome model groups of intestinal mucosa with ELISAmethod. And flow cytometry was used to detect the positive cell percentage oflymphocyte (CD3) in normal control group and other spleen asthenia syndrome modelgroups of intestinal mucosa.Results:1. Compared with the normal group, the secretion of sIgA of the spleen astheniasyndrome model group of TCM, the natural rehabilitation model group,and theSijunzi Decoction control group were decreased (P<0.01), has statisticallysignificant difference. With the therapeutically treatment of SijunziDecoction, secretion of sIgA of the Sijunzi Decoction control group increasedbetter than the spleen asthenia syndrome model group of TCM and the naturalrehabilitation model group, has statistically significant difference,(P<0.01or P<0.05).2. Compared with the normal control group, the positive cell percentage oflymphocyte (CD3) of the spleen asthenia syndrome model group of TCM, thenatural rehabilitation model group,and the Sijunzi Decoction control groupwere decreased (P<0.01), has statistically significant difference. Withthe therapeutically treatment of Sijunzi Decoction, positive cell percentage of lymphocyte (CD3) of the Sijunzi Decoction control group increased betterthan the spleen asthenia syndrome model group of TCM and the naturalrehabilitation model group, has statistically significant difference,(P<0.01).Conclusion: In the state of spleen asthenia syndrome, the secretion of sIgAdecreased, and the positive cell percentage of CD3lymphocyte decreased. Theimmune barrier function of intestinal mucosa is disorder. While SijunziDecoction can improve immune ability of the rats.
