Objective:This research is under the instruction of preventive treatment of disease. The main research contents are emotional factors, and regulating mood is the main therapeutic method. This research aims to discuss the effect of relieving qi stagnancy in Liver to emotion caused breast diseases and its mechanism.Methods:The SPF degreed C3H/HeJ female rats were divided into 4 groups at random: blank group, model group, Chinese medicine group, western medicine group. From 1 to 10 week, model group, Chinese medicine group and western medicine group are intervened with chronic multiple compound stress; blank group without intervention. From 11 to 14 week, blank group was given gastric perfusion of distilled water; model group was intervened with chronic multiple compound stress and gastric perfusion of distilled water, Chinese medicine group was intervened with chronic multiple compound stress and gastric perfusion of Shugan granule; western medicine group was intervened with chronic multiple compound stress and gastric perfusion of tamoxifen.We observed and recorded the biological characterization of changes at week 0,10,14. After the experiment, we stripped the hypothalamus neurotransmitters, took blood from eyeballs to detect serum hormone, and took mammary gland and its surrounding skin aseptically for pathological observation and immunohistochemical method to detect tissue ER、PR、Ki-67、P63.Results:After the modeling at week 10, model group, Chinese medicine group and western medicine group showed a series of biological characterization of changes compared with the blank group:fur dim, plucking, and curled up cluster rested and squinting, listless apathy. Among the three groups, the incidence of alopecia is over 80%. Nose was the starting point of hair removal and diffused toward the back, which can reach eyes. Some were with beard off, molt can be found at back, abdomen, and limbs in some serious cases. Compared with the blank group, the incidence of alopecia was statistically significant (P<0.017). Tail suspension experiment was to test animal behavior change, results showed that the struggling state time of blank group was obviously longer than the other three groups (P<0.05). The fixed time of blank group was obviously shorter than the other three groups (P<0.05). These indicated that blank group rats were more active, the other three groups showed despair of behavior change after chronic multiple compound stress.After the drug intervention at week 14, several rats mammary bump is obvious through macroscopic observation. There are differences between the four groups, the incidence of western medicine group of mice mammary bump is higher. Under the microscopic breast tissue pathological observation, the three groups showed different degrees of acinar number increase, duct dilatation, excretion in duct, increased cell layer and cells concentrated apart from the blank group. The modeling group is statistically significant compared with the blank group. The hyperplasia of model group was significantly higher than that of the blank group, it suggested that chronic unpredictability psychological stress can increase the incidence of C3H/HeJ female rats mammary gland hyperplasia. The Chinese medicine group is statistically significant compared with the modeling group, Chinese medicine group showed low degree of hyperplasia. This indicated that in the process of chronic psychological stress intervention, the liver granule can reduce the incidence of hyperplasia of mammary glands.In this research, the ER and PR of C3H/HeJ female breast tissue were of negative expression, while Ki-67 is of high expression. This can lead to breast cell proliferation, which may be the cause of C3H/HeJ female rats spontaneous breast cancer.After drug intervention, hypothalamus norepinephrine (NE) and serotonin (5-HT) were less in Chinese medicine group. There was no significant difference between the other three groups. This showed that Shugan granule can suppress the secretion of NE and 5-HT in the hypothalamus. Serum adrenocorticotropic hormone releasing hormone (CRH) and adrenocorticotropic hormone (ACTH) were increased, which is consistent with the Chinese medicine group in neurotransmitter test results, it was because NE can inhibit the secretion of ACTH. Serum gonadotrophin releasing hormone (GnRH) content detection showed that traditional Chinese medicine group is higher than the other three groups. Model group was higher than the blank group and western medicine group. Chinese traditional medicine group of estradiol (E2) and progesterone (Pg) is higher than the other three groups. This consistent with 5-HT test results in the hypothalamus. The hypothalamus 5-HT of Chinese medicine group was in a low level.5-HT mainly has inhibitory effect on gonadotropin secretion.Conclusions:This research intervenes through multiple compound chronic stress on C3H/HeJ female rats to simulate emotion causing disorder, which induced spontaneous alopecia on C3H/HeJ female rats. Biological characterization and behavior has an obvious change, proving the success of emotion causing disorder.Through the observation of chronic compound stress effects on mice mammary gland, model group compared with the blank group, there were transformation of gland bubble populations, duct expansion, lumen with secretion, cell layer increased, hyperplasia of cell density change, proving that chronic compound stress has obvious stimulation on mammary gland. Chinese medicine group enjoyed low degree of hyperplasia, it suggested that in the process of intervention of chronic psychological stress, the liver granule can reduce the incidence of hyperplasia of mammary glands. From neuroendocrine changes it proved that Shugan granule can affect the hypothalamus neurotransmitter, and then by adjusting the hypothalamus-pituitary-adrenal axis (HPA) and the hypothalamus-pituitary-ovarian axis (HPO) to affect serum hormone levels.In this research, the ER and PR of C3H/HeJ female breast tissue were of negative expression, while Ki-67 is of high expression. This can lead to breast cell proliferation, which may be the cause of C3H/HeJ female mice spontaneous breast cancer. The C3H/HeJ female mice breast tissue estrogen receptor was of negative expression, this is different from the human estrogen receptor expression. Human mammary duct and lobular epithelial cells visible ER is of positive cell distribution. Ki-67 are highly expressed in this research. Coincidentally, human menopausal women ER alpha expression and cell proliferation in mammary gland showed a negative correlation between tags. In particular, the majority of ER alpha proliferation related antigen positive cells do not express Ki-67, and Ki-67 positive cells often assumes ER anegative.The incidence of C3H/HeJ female postpartum breast cancer increases, which unlike humans that pregnancy acts a protection for mammary gland. The negative expression of C3H/HeJ mice female progesterone receptor in breast tissue also illustrates the C3H/HeJ female breast cancer pathogenesis is different from humans.
