Investigating The Molecular Mechanism Of Treating Lumbago By Needling

Waist pain is a very common symptom .which usually be called low back pain . It can appear in many disease, such as lumbar interveretebral disc syndrome,lumbar spinal stenosis, myofascial disorders, the inflammation of the lumbar zygapophyseal joint capsules and trauma . Low back pain usually be divided to cold- dampness , dampness-heat, deficiency of kidney and blood stasis in Chinese medicine.Low back pain can be treated by surgery, acupuncture, or with medicine. Acupuncture has many advantage to western medicine, such as no side-effect,no expensive, high effective, ect. Many precursor provided much clinic experience, Many doctors had studied different points and different manual in treating low back pain, but it is notenough to study the mechanism of treating low back pain by acupuncture. Up to now ,we still not take full advantage of modern experiment measure to study this mechanism. We created a lumbago model of blood stasis by trauma in Wistar rat,and treated these rat by acupuncture. Shenyu and Dachangshu had been chosen main points, Weizhong had been chosen minor point. All rats were been randomly divided into three group,A: acupuncture Shenyu,Dachangshu and Weizhong,B:only acupuncture Ashi, C: group model ,D: normal control. Rats were killed in 1 hour or 1 week later. All samples were tests the congestion and apoptosis of lumbar muscle. Brain was test NOS ( nitric oxide synthase ) and c-fos gene protein.Results: The congestion of lumbar muscle in group A and B were lesser than C.The NOS and c-fos expression in CNS were higher in group A than in group B an C. At same time,the expression of NOS and c-fos in CNS was higher in group B and C than group D,but there was no different in expression of NOS and c-fos between group B and C.These results suggest:l:There was abirritation by needling Shenyu, Dachangshu, Weizhong or Ashi point.2:Treating lumbago by by needling Shenyu, Dachangshu Weizhong or Ashi point was provided with relaxing muscle and tendon and activating collateral , or promoting blood circulation to remove blood stasis.3: Relaxing muscle and tendon and activating collateral ,or promoting blood circulation to remove blood stasis were proved by lesser congestion and apoptosis.4.Needling Shenyu, Weizhong and Ashi points posses two different effect,directly abirritation and indirectly abirritation , but Ashi only posses directly abirritation.5. Direct abirritation mechanism may be caused by affecting the NOS or c-fos expression.
