The Experimental Study of repairing meniscus lesion with fibrochondrocyte-collagen constructObjective: To repair the menisci lesion with fibrochondrocyte-collagen construct in dogs.Methods: 1 .To extricate fibrochondrocyte from canie menisci,then expand in vitro, study the biologic behavior; 2.To synthesize collagen-GAG template and fibrochondrocyte were added into the porous collagen -GAG template forming cell-collagen construct, then the cell-collagen construct were implanted subcutaneously into nude mice.After 3,6,12 weeks postoperation ,the implanted collagen construct were evaluated by gross.histology and ultrastructure observations. Finally, the cell-collagen construct were implanted into the meniscal lesion in dogs. 3,6,12 weeks later, the implants were evaluated by gross, histological observations.Results: 1 .Fibrochondrocyte grow ,proliferate vigorously in vitro; 2. Fibrochondrocyte can form meniscus-like tissue in vivo, collagen template was degradable, intoxic and implantable;3.Grossly, histologically, the fibrochondrocyte-collagen implants underwent inflammation and degradation , consequently formed a meniscus-like fibrocartilagetissue. Histologic staining showing the repair tissue of the cell-collagen group was of fibrochondrocyte tissue which resembled the ordinary fibrochondrocyte very much; the repair tissue of the collagen group was like fiber tissue ,and that of the vacant control group was only partial scar tissue. Conclusion:!.The fibrochondrocyte can be extricated from meniscus and expanded in vitro; 2.Fibrochondrocyte could grow and synthesize extracellular matrix in collagen construct ;4.The meniscus lesion could be excellently repaired with the fibrochondrocyte-collagen construct.
