Part one: The dynamical changes of nitric oxide (NO) in the blood detected by ESR during myocardial ischemia-eperfusion injury in rabbits. Objective: In order to offer the direct evidence of the dynamical changes of nitric oxide (NO) in the blood detected by ESR during myocardial ischemia-eperfusion injury (MIRI) in rabbits, Two methods were tried in this part. Study 1 Objective: Spin trapping of NO inside the body. Methods: The rabbits were anesthetized with intravenous pentobarbital(4Omg/kg). DETC(500mg/kg)were dissolved in normal saline, and FeSO4(5Omg/kg) plus sodium citrate(250mg/kg), and all they were injected intraperitoneally and subcutaneouslly,respectivelly at the same time. Samples of venous blood were obtained for the detection of NO. Results: The ESR signals were only seen at the time between 20 to 40 minutes after the injection of spin trap, there were no signals after 40 minutes 10 Conclusion This method could not reflect dynamical changes of nitric oxide (NO) in the blood during inyocardial ischemia-eperfusion injury, so it was given up. Study 2 Objective: Spin trapping of NO outside the body. Methods: DETC( 6 mmol/L) and FeSO4 (3 mmol/L) were dissolved in normal saline respectively. 20u1 of the FeSO4 and DETC were immediately added into the samples of venous blood (lml) after taken separately, and then it was put into the 37t water pool for 10?5 minutes. After that 0. 4m1 ethyl acetate were added with shaking which was centrifuged at 12000 rpm. The extraction was used for ESR detection of NO. Results: The ESR signals were stable at different times. Conclusion: This method could reflect dynamical changes of nitric oxide (NO) in the blood during(MIRJ), so it could be taken in this part. Study 3 Objective: To observe the dynamical changes of NO level in the blood during MIRI in rabbits Methods: MIRI model was made in five of New Zealand White Rabbits, the left anterior descending coronary artery(LAD) was occluded for 10 mm prior to 120 mm reperfusion. NO were detected at the time of before the ischemia(baseline), 5, 10mm during the ischemia and 5, 10, 30, 60, 90, 120mm after the reperfusion, and dp/dtmax were recorded at the same time. 11 Results NO level were elevated significantly at 5min(23 4?.8mm) after the reperfusion vs baseline (12.0?.5mm) p
