Objective To prove that the p16 raethylation occurs in theearly stage of lung cancer by studying the p16 methylationstatus of BEP2D cell during its malignant transformation.Method Normal BEP2D cell , BEP2D cells irradiated by aparticle for 20 weeks(R-20),30 weeks(R-30), 35 weeks(T-35) and54 weeks (T-54) were chosed to study the p16 methylation statuswith the method of MS-PCR(methylation-specif ic PCR), meanwhile,RT-PCR were used to study p16 mRNA expression of these cellsto make sure that the methylation of p16 causes the inactivationof p16 protein expression.Result 1. No p16 methylation were found in BEP2D cell, p16methylation were found in R-20 , R-30, T-35, T-54 cells. 2. Thep16 protein expression of R-20, R-30, T-35, T-54 cells weremuch lower than that of BEP2D cell.Conclusion 1. The p16 methylation occurs in the early stageof lung cancer .2. The methylation of p16 causes theinactivation of p16 protein expression.
