Gestational trophocyte is a very special kind cell, as a semi-heterogeneous cell originated from embryo, not only could successfully escape from immunosurveillance and immunologic rejection of mother's, but also posses some malignant biological behave such as invasion and is similar to tumor in morphology in some degree, with some tumor antigens expression. Trophocyte transfer from chorial surface to mother decidual stroma, replacing arteriole wall existed in mother's coiled arterioles that is necessary for trophoblast undertaking normal function which needed in accurate control while not existed in other benign cells, or it can't realize truly embedding. Secretion human chorionic gonadotrophin(hCG) is one of the feature of trophoblastic cells.In physiology, hCG secreted by placenta plays an important role in pregnancy, by promoting menstrual corpus luteum developing to gestational corpus luteum which secrete enough progesterone to keep normal pregnancy. In pathology, hCG can be synthesized and secreted by trophoblastic tumor cells,that being identified as the tumor marker of trophoblastic tumor clinically.hCG is a characteristic hormone secreted by trophoblastic cells. Secretion hCG is the feature of trophoblastic tumor. hCG is a marker for diagnosis and treatment monitor for trophoblasic tumor. The level of hCG up and down refracts the situation of normal gestational and suggests trophoblastic tumor cell's increase or decrease. However, there are few study on hCG act on itself secreting cells?trophocytes. It is still obscure whether hCG influence biologic function, endocrine, cellular growth and development of the trophoblast. If inhibiting the function of trophoblastic tumor secreting hCG could bring some changes in morphology and regulate it's growth or induce it's apoptose. In whole, It is still a question required study whether hCG participate in the functional regulation in trophoblastic growth and increasing acting as a autocrine hormone, how about it's acting in tumor cellular proliferating and invasion.Thus, making study in light of hCG secretion, investigating it's autocrine may plays a significant role in study of trophocyte regulation function, even in treatment of trophblastic tumor and may have a potential practice view.Trophocyte synthesize and secrete hCG, from the view of molecular-biology level, which is a result of hCG gene expression. If synthesis and secretion of hCG be controled or blocked by molecular-biology method, it may be possible to realize studying on hCG autocrine function and observing biological effect induced by absent hCG.In 1984, Izamt and Weintraub first pronounced the concept of "antisense technology". Antisense technology means in light of complementary base pairing theory, using " artificial or biological synthesized special complementary DNA fragment, inhibit or block some genes, making it inactive, then attaining to control gene expression or treatment of some diseases. After more than ten year's preclinical study, extend using of antisense technology has come into possibility. MethodsThis study chooses choriocarcinoma strain JAR as study subject. Antisense oligodeoxynucleotide(AS-ODN) designed based on hCG P subunit DNA sequence used to inhibit JAR cell to secrete hCG in vitro.Using RT-PCR determine JAR cell and normal gestational trophocyte LH/hCG receptor mRNA; Observing the changes of endocrine function of secretion estradiol,pregesterone and hPL by JAR cells, it's growth situation, cellular proliferation cycle or it happen to apoptose when hCG was inhibited or added external hCG in vitro. In test, using sense oligodeoxynucleotide(S-ODN) and random sequence oligodeoxynucleotide(R-ODN) as controls exclude nonspesific action, taking hCG secretion negative cell acts as negative contrast group. Results1. Compared with contrast group, after AS-ODN 5umol/L,10umol/L and 20nmol/L act on JAR cell 24hr, hCG secretion reduced 23.9 percent (p<0.01),57.4 percent (pO.Ol) and 74.4percent (p<0.0Irrespectively.
