Experimental Autoimmune Myasthenia Gravis in RabbitInduced by Tal25-147 and Tal83-200Objective To induce experimental autoimmune myasthenia gravis (EAMG) rabbits for the study of therapy on myasthenia gravis (MG). Methods Peptides Tal25-147 and Tal83-200 were synthesized referring to the residue sequence of acetylcholine receptor of Torpedo California and the rabbits were inoculated with the peptides respectively to make Tal25-147 immunogenic group and Tal83-200 immunogenic group. Clinical manifestation was graded in 4 levels. Electrophysiological function was assessed by repetitive nerve stimulation (RNS) and single fibre electromyography (SFEMG) tests. Anti-peptides antibodies were evaluated by enzyme linked immunosorbent assay (ELISA). Student t test were used to analyze the difference between the EAMG and healthy rabbits, and to find the diversity in the two immunogenic groups. Results The first day after the injection, all the rabbits showed fatigability, but recovered quickly. The body temperature rose up mildly and fell to normal in not more than three days. Followed the booster, the rabbits appeared weakness, and Tal25-147 group developed quickly. Clinical symptoms could be improved by neostigmine. At 3 , 5 > lOHz, the decrement of compound muscle action potential (CMAP) and the mean jitter (MCD) of all the immunized rabbits were higher than the healthy one. The percentage of decrement of CMAP in the order of control group, Tal25-147 group, Tal83-200 group were: 3Hz : 1.625 + 1.317* 25.375 + 7.945 , 20.500 ?9.180 , 5Hz : 2.000+1.732, 25.625 + 9.102, 24.000 + 9.024, lOHz: 1.750?1.392, 28.875 + 8.709, 27.000 + 8.880. Following the above sequence, the MCD were 3Hz: 9.875+1.126, 25.875 + 7.945, 20.000 + 4.260, 5Hz: 11.375 + 0.916, 27.500 + 3.381, 21.375 + 1.685, lOHz: 12.375 + 1.061, 31.000 + 4.811, 24.250+1.188. In the Tal25-10147 group, the decrement at 3 Hz, and the MCD at 3 > 5 > lOHz were more severe than that in the Tal83-200 group. Anti-peptide antibodies in immunized groups were significantly higher than that of the control group. The positive ratio of anti-peptide antibody in Tal25-147 group was 8/8, and that in Tal83-200 group was 7/8.Conclusion Tal25-147 and Ta 183-200 were immunogenic in the induction of EAMG in rabbits, accompanied by elevation of anti-peptide antibodies, blockage of neuromuscular transmission, neostigamine could improve the manifestation of the rabbits...
