Renal interstitial fibrosis is inevitable pathologic change in chronic renal disease inducing to renal failure. A lot of studies and clinical observations have shown that the tubulointerstitial injury play more dangerous role than the glomerular damage in the kidney . To study the mechanisms of renal interstitial fibrosis and the treatment-resistant of drug may be beneficial to the options of drug in clinical treatment. Chinese herbs in activating the blood circulation and removing the blood stasis , such as Danshen parenteral solution (salvia miltiorrhiza compound parenteral solution) and Bloodsucker are commonly used in kidney disease but the roles of them on antagonist interstitial fibrosis are still unknown. So the rats with unilateral ureteral obstruction(UUO) were adopted to result in renal interstitial fibrosis and were used to study the effects of Danshen parenteral solution -. Bloodsucker and benazeprol(angiotensin converting enzyme inhibitor). Angiotensin converting enzyme (ACE), Angiotensin II(Ang II), endothelin-l(ET-l), transforming growth factor-6, (TGF-B,), nitric oxide (NO) and collagen IV(coll IV) were observed in the experiment. Isolated perfused kidney (IPK) was used to estimate the function of obstructed kidney. Apoptosis was also observed by TUNEL. Through clinical observation, Chinese herbs were used in patients of chronic nephritis to protecting the renal function. Results were as follows: 1. The roles of Chinese herbs on the vascular active matter in renal tissue of experimental rats with obstructive nephropathy.The vascular active matter such as Ang II and ET-1 can contract the blood vessel and accelerate the progress of renal interstitial fibrosis. WhileNO can expand the blood vessel and postpone the progress of renal interstitial fibrosis . 70 healty female six week old SD rats were randomly divided into 7 groups (n=10) ,which were Sham group, UUO group, UUOD group (treated with Danshen, 3 g/kg/day), UUOB group (treated with Bloodsucker, 3 g/kg/day), UUOL (Benazepril, 4 mg/kg/day), UUODL(Danshen plus Benazepril), UUOBL (Bloodsucker plus Benazepril). They all underwent left unilateral ureteral obstruction (UUO) or sham operation. Except Sham group , the other groups were given drugs by oral one day before operation. The rats took the water and food freely until put to death 5 days after the operation. The kidney of operated side was selected. Each part of the kidney tissues were homogenated for detecting the content of ACE. Angll > ET-1 and NO by radio-immunity method and chromatometry. And part of the renal tissues were fixed in formalin, embedded in paraffin and made into sections to observe constructive nitricoxide synthase (cNOS) by immunohistochemical method. Renal tubule and epithelial cells were also observed by HE and Masson staining.1.1 ACE and Ang II in renal tissues : ACE in UUO group (32.8 ?.3u /mg protein) was higher than that in Sham group(13.1?.2 pO.OOl); ACE in UUOD group(13.3?.8), UUOB group (14.2?.7) and UUODL group (11.8?.0) were less than that in UUO group (pO.OOl); ACE in UUOL group (16.6?.6) and UUOBL group (14.2?.4) were lower than that in UUO group (p<0.01). Ang II in UUO group (80.9?7.3) rose markedly compared with Sham group (15.3?.1 p<0.001pg/mg protein); in treatment groups such as UUOD group (42.0?3.9), UUODL group (29.U8.7), UUOL group (32.5?.1) compared with UUO group, Ang II reduced significantly (p<0.001); it also reduced in UUOB and UUOBL groups (40.8?6.0, 35.6?6.9 p<0.01).1.2 The detection of the content of ET-1 and NO in renal tissue: ET-1 in UUO group (250.67?9.80 pg/mg protein) was significantly higher than that in Sham group (77.59?8.30 pO.OOl). In UUOD, UUODL andUUOBL groups(161.79?3.85, 160.28?9.97, 158.03?5.63) ET-1 was inhibited. It was much lower than that in UUO group(p<0.05). In UUOL group(143.58?7.42) ET-1 was lower than that in UUO group (p<0.01). There is no significant difference between UUO and UUOB group (186.65?9.93 p>0.05).
