Osteoporosis has become a big problem in social hygiene in ageing society and is paid more and more attention by the global medicine circle, mainly because of fracture and pain resulting from osteoporosis, primarily forearm fracture, lumbar fracture and hip fracture. In which hip fracture is the most serious, with high incidence of a disease and death rate, as well as high treatment expenses. Thus, the living quantity of the elderly people is seriously influenced by osteoporosis. China has entered into ageing society since 1998, with 1/5 ageing population of the world and 1/2 of Asia, thus, vast medical resources will be expended on osteoporosis. In most countries, the incidence of osteoporosis of the female is about 2-4 times of that of the male, this is type I congenital osteoporosis which results from the decrease of estrogen after menopause, which is also the research focus of in this dissertation.Summarizing the theories of the doctors of the past dynasties, Chinese Medicine's cognition of the pathogeny and mechanism of osteoporosis is that, deficiency of the kidney results from all kinds of causes, characterized by essence deficiency, especially kidney deficiency as well as liver deficiency, lack of qi of spleen, qi and blood. Most literature considers that deficiency of kidney and spleen are the main pathologic factor, while clinic varies with personal born gift, there are different symptom.Jian-Gu-Er-Xian extraction is expurgated from Gui-Lu-Er-Xian colloid, which was created in Ming Dynasty. Added some Chinese herb, Jian-Gu-Er-Xian extraction becomes more effective in supplementing the spleen and boosting Qi. Based on the cognition of the mechanism of osteoporosis of the Chinese Medicine and research done before, this dissertation studied effect pharmacological mechanism of Jian-Gu-Er-Xian extraction on osteoporosis.This experiment studied bone biomechanics, histomorphometric analysis and proteomics with ovariotomy (OVX) rats, which simulated postmenopause osteoporosis. Meanwhile, with rats' osteoblasts as material, external research was done to study effect of Jian-Gu-Er-Xian extraction on cell activity, PKC, expression of anti-apoptosis protein Bag-1 and relation between its effect approach and estrogen receptor.From the results, we know:1. Jian-Gu-Er-Xian extraction can effectively ameliorate the capability of biology mechanics of bone tissue of OVX white rats. In addition, it can prevent bone quantity from losing, ameliorate bone structure. Its effect on ameliorating bone structure and some indicators of biology mechanics is better than Premarin.2. Jian-Gu-Er-Xian extraction can decrease the high transition rate of bone of white rat whose ovary is cut and prevent bone quantity from losing. But it dose not take effect by estrogen. It has differentprocess approach with Preamrin and so on.3. The change of expression of estrogen receptor p and vitamin receptor take part in the osteoporosis occurrence and mechanism of Jian-Gu-Er-Xian extraction.4. The approach of PKC signal transferring is important in the mechanism of osteoporosis occurrence and mechanism of effect of Jian-Gu-Er-Xian extraction. Jian-Gu-Er-Xian extraction can influence PKC signal approach by increasing expression PKC and PKC-banding protein.5. Jian-Gu-Er-Xian extraction can restrain osteoblast apoptosis. It is one of the mechanisms of preventing osteoporosis. It is one of practical mechanisms of anti-osteoblast apoptosisto Increase the expression of bag-1, so influence ratio of Bcl-2/Bax.6. angiopoietin-related protein 3 , GABA A receptor alpha, PEST phosphatase interaction protein, NFkB essential modulator, nay modulate the occurring and therapy of osteoporosis, too.7. From the literature research, it can be conclude that this experiment takes the lead in introducing the technique of proteomics in modern research of Chinese medicine as well as orthopedics.
