Objective:To investigate the effect of acupuncture, moxibustion or total glucosides from cornus officinalis(TGCC) on the bone metabolism index of ovariectomized rats by using bone histomorphometric method, a comparative study was undertaken. And a preliminary study on the mechanism of acupuncture preventing and treating osteoporosis was made, so tc offer an experimental basis for clinical spread and application of using acupuncture and moxibustion to prevent and treat osteoporosis as well as to find an effective way to prevent and treat osteoporosis. Method1. Animals and groups: seventy 6-month old female Wistar rats were divided randomly into seven groups: sham-OVX, model, acupuncture, moxibustion , high -dose of TGCC, middle-dose of TGCC and low-dose of TGCC. 10 rats in each group.2.Models:Except the sham-OVX group, all the models were produced by bilateral ovariectomy. Then all the animals were fed under the same condition(room temperaturel9~24℃,relative humidity 40% around) with usual feedstuff and allowed free access to water .3 .Acupuncture and medicine: From the 4th day after the operation, the acupuncture group was treated with puncturing bilateral "shenshu" and "sanyinjiao" acupoints with reinforcing method by twist, lmin for 1 acupoint and moxibustion group with moxibustion on the same acupoints,5min for 1 acupoint. All the treatment lasted for 3 months, 6 times a week.The dose given by gastrotube of total glucosides from cornus officinalis(TGCC) were converted by the ratio of surface of rats to that of human.The concentration is separately 3.56g·kg-1(1.6 time of equivalent dose), 2.16 g·kg-1 (1 time of equivalent dose).1.08 g·kg-1 (0.5 time of equivalent dose). 8ml for each time, once a day,6 times a week, for 3 months.4. The labels of tetracycline: All the rats were give injection of tetracycline30mg / kg twice on the 10th day and 1th day before killed. The right distal femur were processed undercalcified sections for quantitative bone histomorphometry.5.Observation method: Three areas of bone trabecula under the epiphysial cartilage on the undercalcified sections were selected to input the computer. Each section for 3 kinds of Observation method and 3 images for each kind(Histological observation, under Gemisa staining and usual light source; Osteoid observation, under vonkossa staining and usual light source; Mineralization observation under fluorescence). The images were measured by bone histomorphometric method with Image-Pro plus soft ware of American Pixera6000CL instrument of computerized image analysis, then the data were disposed by statistical method with SPSS software. Result:1 .After the operation, the bone trabecula under the epiphysial cartilage of rats in model group were significantly less and thinner, the bone absorption was enhanced, the osteoid was significantly more than the sham group. Under fluorescence, the mineralizing surface was enhanced. Osteoblast surface, osteoclast surface.osteoid surface and osteoid width were significantly improved than the sham group. The mineralization deposition ratio, and ajusted mineralization deposition ratio were enhanced. The mineralization lag time was shorter. All of this proved that OVX could induce osteoporosis of high bone transformation .2.After treatment of acupuncture,moxibustion and different concentration of TGCC, the bone trabecula surface, osteoblast surface were increased, osteoclast surface was decreased of all groups, demonstrating in all groups, the improvement of bone loss existed. The acupuncture group and middle-dose of TGCC group could promote the bone formation significantly. The acupuncture group could inhibit the bone absorption too.3. After treatment of acupuncture, moxibustion and different concentration of TGCC, the mineralizing surface decreased and the distance of double label were narrowed, demonstrating the bone transformation decreased, bone remodeling time...
