Part I Culture ex vivo and identify of dendritic cell (DC) Objective To investigate dendritic cells of peripheral blood of patient with hepatocellular carcinoma compared with that of normal donor. Methods To establish the culture system ex vivo of human dendritic cells and to analysis the phenotype of DCs,Results 1 .Established successfully the culture system of DC ex vivo . 2. Analyzed the phenotype of DC and identified the DC with FCM. 3. Compared the harvest rate and the purity of DC,no significant difference between patient DC and normal donor DC (p>0.05) 4. There is no significant difference of capacity of activated lymphocyte between patient DC and normal donor DC (p>0.05) 5. The capacity of secreting IL-12 is almost same in patient DC compared with healthy donor DC. Conclusions The ideal DCs both in functionally and quantity can be obtained in peripheral blood of patient with primary hepatocellular carcinoma,and can be used as vaccine for HCC immunotherapy.
