Purpose: To establish a primary culture method of normal person and Thyroid associated ophthalmopathy patients to inform the living of preadipocytes for in -depth studing the function of preadipocytes in TAO.Methods: Fat tissue culture method was used to culture preadipocytes of normal person and patients with TAO. Normal orbital fat tissue took from atrophy of eyeball patients after artificial globe surgery . Orbital fat tissue took from Patients with TAO after orbitotomy.By observing cell, MTT method was used to describe growth curve and biology performance,flow cytometer to identify preadipocytes.Results: Orbital preadipocytes from normal person and patients with TAO were cultured successfully. The cells purified and cultured were highly homogeneous , highly proliferative and with high differentiation rate .The shape of cells was the same with fibroblast . Orbital preadipocytes patients with TAO were more proliferative than preadipocytes from normal person ,the shape of cells was the same.The primary culture of two preadipocytes grew slow,and grew guicker in the second and third generations .After 3-5days of primary culture ,we could observe cells swim from tissues.First generate after 10 days.The growth curve was also the same.The fifth day entered log phase,The ninth day enter plateau.Human adipose-devived stromal stem cells sueface protein CD44 and CD29 are positive .Use flow cystomic dentify preadipocytes,and identify preadipocytes in patients with TAO orbital tissue.Conclusions:Preadipocytes existe in orbit fat tissue of both in normal person and patients with TAO and have not many differences in bionomics.The preadipocytes act very important role in mechanism of TAO.
