Objectives: To compare the different effect of XIFENGTONGLUO and YIQIHUOXUE on apoptosis of continuously ischemic gerbil brain. To compare the different effect of XIFENGTONGLUO and YIQIHUOXUE on apoptosis and apoptotic adjustive gene bcl-2 and bax expression in continuously ischemic rats brain. Methods: Ligating gerbil right carotid to make the model of continuously cerebral ischemia, then detecting the apoptosis in post-ischemic right brain by TUNEL. Giving high lipids diet to rats for making model with hypercholesterolemia. Then ligating rats' bi-carotid to make the model of continuously cerebral ischemia, and then detect the apoptosis of brain by TUNNEL and detect the expression of bcl-2 and bax by RT-PCR. Results: There were some apoptotic cells in right brain posting ischemic 6 hours in control group. There were less apoptotic cells in XIFENGTONGLUO group and YIQIHUOXUE group in gerbils. But there were more less apoplotic cell in XIFENGTONGLUO group than YIQIHUOXUE group. Posting 48 hours of ischemic, most gerbils of the control group were markably more apoptotic cells in brain. In contrast, very less apoptotic cells can be found in brain in XIFENGTONGLUO group and YIQIHUOXUE group, and much more apoptotic cells can be found in brain in XIFENGTONGLUO group(P<0.05). Markably more apoptotic cells could be found in posting ischemic 24 hours in control group rats. Less apoptotic cells could be found in XIFENGTONGLUO group and YIQIHUOXUE group. But there were more less apoptotic cells in XIFENGTONGLUO group than YIQIHUOXUE group. The exprssion of bcl-2 was low in the control group and the expression of bax was high in the control group. In contrast, the expression of bcl-2 was the highest in XIFENGTONGLUO among these five groups and the expression of bax was lower in XIFENGTONGLUO group than in the control group(p<0.01). Conclusion: Treating with XIFENGTONGLUO can suppress the apoptosis in the gerbils' brain, and it is significant than treating with YIQIHUOXUE. It is surely that the method of XIFENGTONGLUO have the function of suppressing the apoptotic geneexpression and activating the suppressing apoptotic gene expression. It is significantly different than treating of YIQIHUOXUE.
