ObjectiveAdenoid cystic carcinoma is one of the most common maxillofacial malignant tumor. It is 9.95% of salivary tumor, 24% of malignant salivary tumor, and the second of malignant salivary tumor. Adenoid cystic carcinoma often happens in small salivary gland and smaller gland of big salivary. It occurs in head and neck, and is rare in other part of body. According to WHO, there are 3 types; adenoid type, tubular type and parenchymal type. The three types can happen in one tumor. According to the ratio of constituent, the tumor is typed. It seems that there is clear boundary between adenoid cystic carcinoma and peripheral tissue in salivary gland. But there is no envelope around the tumor, and the tumor often infiltrates peripheral tissue. In a few of cases, there is incomplete envelope around the tumor. Adenoid cystic carcinoma is a kind of infiltra-tive tumor, and tumor cell can be found in and out of envelope. Near the tumor, such as muscle, bone and periosteum, the infiltrative tumor cells, which cannot be found by eye, can be tested under the microscope. So it is difficult to judge the anatomic field of tumor. The rate of distant metastasis is very high. Early metastasis can occur, and lung is the main part involved. Recovery can occur after operation. So it is difficult to treat and it does harm to health of patients. Control of recovery and distant metastasis is an important question in clinical treatment, and now research of pulmonary metastasis of adenoid cystic carcinoma is a focus.Protein kinase is one of family of enzyme, and a catalyst of protein phos-phorylation, which is a common mechanism of cellular action. CK2 is the mosteffective protein kinase. CK2 is consist of two catalytic subunit(α,42 -44KDa and α ' ,38KDa)and two regulatory subunit( β,28KDa) , and there are three types: ααββ, αα 'ββ andα ' α ' ββ. The structure is stable, and only decomposed in vitro. Subunit p plays a role in expression of activity and adjustment of substrate. Up to now, more than 160 substrates of CK2 have been found. The substrates involve in regulation of transcription, signal conduction, regulation of growth and development and structure of cell. Except cellar matrix, some virus protein can be phosphorylated by protein kinase CK2. Protein kinase CK2 plays a role in transcription, signal, growth and development of tumor.CK2 can phsphorylate many substrates which involve in cellar fission and differentiation and development of tumor. Through immunohistochemical method expression of protein kinase CK2 - p in SACC is evaluated to investigate significance of interaction of protein kinase CK2 - p in SACC. To study the relationship of protein kinase CK2 - p and pulmonary metastasis, we extract nucleus and plasma of SACC - LM and SACC - 83, evaluate activity of protein kinase CK2 - βand expression of protein, and disclose the influence of protein kinase CK2 - β to pulmonary metastasis in protein level. Nm23 - H1 is a kind of inhibitory gene of tumor metastasis. Its expression is related with metastasis of human malignant tumor, and it is confirmed that it is negative correlation with pulmonary metastasis of adenoid cystic carcinoma. The fact that protein kinase CK2 -β is related with pulmonary metastasis of adenoid cystic carcinoma has not been known. In the experiment, DRB, an inhibitor of CK2 - β, is used to manage SACC - LM, and then mRNA of nm23 - H1 is evaluated to investigate the influence of protein kinase CK2 -β to pulmonary metastasis of adenoid cystic carcinoma.Method1. immunohistochemical methodThere are 45 cases of SACC and 12 cases of normal salivary tissue, which have been identificated and tpyed by two pathological professors. The fresh tis-sue was fixed by 10% formaldehyde solution, and 5|xm paraffin sections were made.
