High intensity focused ultrasound (HIFU) is one of the fast developing non-invasive techniques for tumor therapy in recent years, this technology involves multiple technology items such as medical science, biology medical engineering, acoustics, automation control, computer and precise machinery manufacturing etc. It's main characteristic is that with high energy capacity focusing in the focal place, it can cause the tumor tissue coagulation necrosis instantly, without injurying the surrounding tissues and tissues that the ultrasound beam pass through. Multiple experiments and clinical researches indicate that it is effective, safe and feasible for HIFU to treat the patients with liver cancer, breast cancer, osteosarcoma and soft tissue sarcoma etc. In this subject we built lung VX2 tumor animal models, and treated the models (early stage and late stage) by high intensity ultrasound, in order to discuss the effectiveness, safetyand feasibility of HIU to lung cancer. With the results of HIU to animal, we also looked forward to observe the condition for HIFU and HIU managing human lung cancer specimens. All that we did could provide an experimental foundation for clinical application of HIU. From the experiment we have got some results as follows: Chongqing HIFU Company conducted 1.First of all, the HIU therapy system. It has many features such as small volume, lightweight, easy serviceability and extended purpose. It can achieve different objectives with different transducer. And its chief capability is: output power is 0-50W;freqency is 2-10MHz. 2.In order to have this experiment, we divided 40 New Zealand rabbits into 3 groups, and built lung tumor model by three methods: tumor cells solution injection, tumor blocks embedding with or without drainage. Observing the tumor biology property, comparisions are made to find out which is the best of the three methods. Conclusion: the animal model built by tumor blocks without drainage had strong stability, and the metastasis mode was similar to human's lung cancer metastasis mode. From above, it is easy to observe the curative effect for HIU treatment.3. Discussing the effectiveness and safety of rabbit early stage lung cancer treated by HIU. We randomly divided 35 rabbits that were embedded 4 days into HIU treatment group (n=15), surgical operationgroup (n=10) and control group (n=10), observing the living quality, existence time, cure rate and changes of weight. Randomly selected 5 rabbits from HIU group and put them to death for pathology and electron micrograph observing; Randomly selected 10 rabbits from HIU group, draw blood from ear artery to make analysis before HIU and 14 days after HIU treatment. Conclusions: rabbits of HIU and surgical operation group existed more than 150 days, longer than the control group (P<0.05), thus we can say that they were cured, and the two groups had no significant difference according to the cure rate and averrage existing time (P>0.05); From pathology and electron micrograph observation, we found that tumor cells in the target necrosis, and the cells' nuclear broke and dissolved, the cell form was not complete, cell organs destroyed completely inside the cell, we could not find any tumor cell metastasis in lymph node; The arterial blood gas analysis shown that PO2, PCO2 did not have changes before and after HIU, this demonstrates that it is safe for lung cancer treated by HIU. 4. Discussing the effectiveness of rabbit late stage lung cancer treated by HIU. We randomly divided 45 rabbits that had been embedded 21 days into HIU group (n=15), chemotherapy group (n=10), HIU and chemotherapy group (n=10) and control group (n=10). Comparsions are made as to the rabbits' existence time of four groups. Randomly selected 5rabbits from HIU group and put them to death to have pathology and electron micrograph observation.
