High intensity ultrasound is a kind of new technology which can remotely destroyed deep-seated tumor tissue without causing damage to the intervening tissue. An ultrasound beam emitted from a focused transducer can pass through tissue and converge into a small volume of tissues at a predetermined depth (targeted tissue), the ultrasound intensity is much higher in the targeted tissue than that in front of or behind the focal zone. The higher intensity ultrasonic will produce rapid , intense heating and cavitation to cause irreversible lethal damage to the targeted tumor cells. HIU has been used to treat liver cancer , breast cancer, bone sarcoma , et al and identified to be effective. Recently, some people has treated experimental vulvar carcinoma and carcinoma of endometriium by HIU. Many study suggested that HIU can increased the immune functionof body bearing-tumor. Cervical cancer is one of the most common woman malignant neoplasms in the world, the lower immune function of patients is one main cause of tumor metastasis and relapse. The effective of HIU treatment and it's effect on immune function of tumor-bearing body are need to study urgently. We investigate the suppress effect of HIU therapy on the reinoculated U14 cervical cancer and the effective of HIU therapy. In this study, light and electron microscopy, MTT, double-colour flow cytometry, enzyme linked immuno sorbent assay and RT-PCR were used to explore the splenocyte proliferation , splenocyte cytotoxicity and spleen Th1/Th2 cytokine expression pattern in the mice bearing U14 cervical tumor. Meanwhile, we study the function of macrophage in tumor-bearing BALB/c mice. The main results are as follows: 1.U14 cervical carcinoma cell has stable biology characters, the cancer incidence was 100%. 10 days after HIU treatment, every mouse was reinoculated, when the number of the reinoculated U14 cancer cells was 2 ×106 ,The non-tumor rates in HIU group(A), surgical resection group(B) and non-treatment group (C) were 100%, 75%,0% respectively, which were no significantly different between group A and group B but significantly lower in group C(P<0.05). when the number of the reinoculated U14 cancer cells was 2 ×107 ,The non-tumor ratesin group A, group B and group C were 87.5%,25%,0% respectively, the rate in group A was significantly higher than that in group B and C (P<0.05). Histology examination: Immediately after HIU treatment, target cells shrinked and nucleus condensed; Electron microscopy examination: tumor nucleolus breaked up and dissolved with wadding, organelles disappeared and internal and plasma membrane ruptured, this is inreversible necrosis. We think that it is effective treatment for mice cervical cancer by HIU, mice treated by HIU were more resistance to the same tumor than that treated by operation, it suggested that tumor cells killed by HIU played a role as HIU solidified tumor vaccine.2.Splenocyte proliferation was determined using MTT in mice bearing U14 cervical cancer , the A570 values in HIU group(A), surgery resection group(B), no-treatment group(C) and normal group(D) were: 0.412±0.031,0.247±0.055,0.136±0.011,0.263±0.027 respectively. The A570 value in group A was higher than that in group B,C,D P<0.05).Splenocyte cytotoxicity was examed by double-colour flow cytometry,when effective cells /targeted cells=5:1,the death rates in group A,B,C,D were 55.38±8.16%,43.91±5.69%,31.20±7.12%,32.28±5.84% respectively, the death rate in group A was higher than that in group B,C,D(P<0.05). when effective cells /target cells=10:1,thedeath rates in group A,B,C,D were 56.72±6.40%,45.28±6.47%,37.33±7.68%,39.85±8.59%,respectively. the death rate in group A was higher than that in group B,C,D(P<0.05).we think the U14 cervical carcinoma cells solidified by HIU may have the same antigen as U14 cervical carcinoma cells, the solidified carcinoma cell can stimulating splenocyte, accelerates its activation and proliferation, acts as HIU solidified vaccine. 3.
