Objectives: To investigate the mechanism of acupuncture and moxibustion on postmenopausal osteoporosis and provide data for the clinical use of acupuncture and moxibustion.Methods: (1) animal experiment : 86 female S.D rats aged 12 months were randomized divided into operative group and shamoperative group .Three months after operation ,the rats of the operative group were randomly divided into 4 groups:model group, E2 group, acupuncture group ,moxibustion group .There were 5 groups in total including the shamoperative group .The effects of treatment were determined by the measurement of the biochemical indexes of blood and urine (Serum-Ca,Serum-ALP, Serum-BGP, Serum-CT, Serum-PTH-M, Serum-E2; Urine-Ca, Urine-Cr, Urine-HOP), bone density,biomechanics indexes, histomorphological indexes, impression of TGF- P lmRNA,VEGFmRNA,and Fas mRNA in bone, impression of IL-6 mRNA of osteoclasts in bone marrow.(2)clinical studies: 30 female cases of postmenopausal osteoporosis were randomly divided into two groups : acupuncture group and moxibustion group.The cases were treated respectively with acupuncture and moxibustion for 180 days .Observed indexes were as follows: clinical symptoms and signs (spontaneous pain of lumbus ,knees and back, oxycinesis on standing up and squating ,lumbus and back pain on walking ,pain on tapping vertebral body ) ,signs of traditional Chinese medicine ( limp wilting lumbus and knees,deafness and tinnitus, haposexuality,spontaneous sweating or night sweating ) and bone density of L1-L4 and femoral neck.Results: (1) 6 months after ovariectomy, compared with the model group,the levels of Serum-ALP decreased significantly (P0.05), and the levels of Urine-Ca/Cr and Urine-HOP/Cr were lower in the acupuncture group,moxibustion group and E2 group.In the model group,Serum-E2 and Serum-CT dropped significantly,and Serum-PTH-M increased .There were significant distinctions in Serum-E2 ,Serum-CT and Serum-PTH-M between the model group and any other group (P<0.05-0.01).After 3 months by the therapy of the acupuncture,moxibustion or estradiol,the levels of Serum-BGP increased, and the levels of Serum-PTH-M decreased lightly in the acupuncture group,moxibustion group and E2 group.As for Serum-BGP and Serum-PTH-M ,there were no significant distinctions between the 3 groups and model group,andthere were distinctions between the 3 groups and shamoperative group (P0.05). The level of Serum-Ca in every group changed little during the therapy (P>0.05).(2)Compared with the shamoperative group in the same period, bone loss ratio in the model group was 13.7%,5.2% in the E2 group,6.7% in the acupuncture group,and 8.8% in the moxibustion group.(3) 6 months after ovariectomy, the width ratio of bone trabecula to marrow cavity was significantly different between the model group and any other group(P<0.01).The ratioes in the E2,acupuncture group and moxibustion group were close to the shamoperative group (P>0.05).There were no significant distinctions among the acupuncture group,moxibustion group and E2 (P>0.05). (4) The luminosities of TGF- B 1,VEGF in the model group were lower than those in the shamoperative group (P<0.05).Those were higher in the acupuncture group,moxibustion group and E2 group than in the model group (P<0.05).Fas protein luminosities in bone tissue was significantly higher in the model group than in the shamoperative group(P<0.01),and lower in the acupuncture group,moxibustion group and E2 group than in the model group (P<0.05-0.01).The model group was higher than the shamoperative group in positive expression ratio of Fas protein in osteoblasts(P0.0l), and the positive expression ratio of Fas protein in the E2 group was lower than that in the model group (P<0.01);Compared with the shamoperative group,the positive expression ratioes of Fas protein in osteoclasts were significantly higher in the acupuncture group,moxibustion group and E2 group (P<0.05-0.01).(5)In the formation number of osteoclasts,the model group was significantly higher than the shamoperative group ( P<0.01),while the ac...
