Background: To test the biocompatibility of biomaterials is very important in the interventional treatment of congenital heart disease. One of the focuses of biocompatibility study is the adherence and growth of endothelial cells to material. But only a few studies are concerning how to promote the adherence and growth of endothelial cells to material, and none of them has good effect on these aspects.Objective: This study is to detect the adherence and growth of endothelial cells to polyamide, and the biological activity of urocortin on the adherence and growth of endothelial cells to polyamide. Methods: The endothelial cells were cultured in the presence of the polyamide. By immunocytochemistry, the cell adhesion and proliferation were evaluated using the following cell markers: vinculin, fibronectin and Ki-67. To double-label the cell markers in endothelial cells which adhered to the polyamide surface, the following antibodies were used: anti-vinculin, anti-fibronectin, anti-Ki-67, TRITC-goat-anti-mouse IgG,FITC-donkey-anti-rabbit IgG, FITC-goat-anti-mouse IgG. Results:1. There was no significant difference in the number of endothelial cells and the expression of vinculin, fibronectin and Ki-67 in the endothelial cells adhered to polyamide and PTFE.2. The proliferation of endothelial cells in urocortin group at the dose of10-7 M was higher than that at the dose of 10-8 M, 10-6 M and nourocortin. 3. The expression of Ki-67 in the endothelial cells adhered to polyamidein the presence of urocortin was higher than that of without urocortin. Conclusion:1. The biocompatibility of polyamide is good. Endothelial cells can adhere and grow on the polyamide surface, and the activity of adherence and proliferation of cells is good.2. Urocortin can promote the proliferation of endothelial cells, and the activity is related to the dose of urocortin.3. Urocortin promotes the growth of endothelial cells to polyamide, and this could be related to the more proliferation of endothelial cells caused by urocortin.
