INTRODUCTIONPeritoneal metastais is the most common form of postoperative recurrence in patients with gastric cancer. The diffusely distributed lession is not suitable for the surgical resection or radiotherapy. Chemotherapy cann't result in satisfying result either. In the last a few years considerable attention has been paid to the use of biological reponse modifiers as an adjuvant immunotherapy to the treatment and prevention of peritoneal metastasis. There is increasing evidence that milky spots are related with peritoneal metastasis of gastric cancer. They are predominant sites for local tumor cells to proliferate and accumulate in peritoneal cavity. Milky spots, mainly locating in greater omentum, now referred to as o-mentum - associated lymphoid tisssue, are accumulations of leucocytes where macrophages are the predominent cell type. The cytotoxicity of the non - activated macrophages is very weak. The activation of macrophages and increase of its cytotoxity to kill tumor cells spreading to peritoneal cavity are beneficial to the prevention and treatment of peritoneal metastasis of gastric cancer. Shimotsuma found the milky spots and milky spots macrophages were activated, the number of the milky spots was increased significantly, and the morphology of milky spots changed following the ip administration of OK -432 in rats. Koenen, by intro-peritoneal administration of GM - CSF in rats, demonstrated both an enhanced antitumour activity of the milky spot macrophages and increase in the milky spots macrophage population. There are few documents in our country about activating milky spots following ip administration of BMRs.It is an important research topic of the immunotherapy for tumor peritoneal dissemination to apply effective BMRs that activite milky spots and milky spots macrophages and to develop its positive effect in the anti - tumor immunity. In the early stage of the invasion of tumor cells, there is a large quantity of macro-phages in the milky spots that means the E/T ratio is sufficient. How to select effective BMRs with little side effect, activate macrophages, and kill tumor cell is the main target of this study. The present study investigated whether milky spots and peritoneal macrophages were activated and whether macrophages developed cytotoxicity following ip administration of INF - 7, Staphylococcin Au-reus Injection, NDV respectivly, with the aim to set up experimental foundation for the prevention and treatment of the peritoneal dissemination of gastric cancer.MATERIALSExperiment animal; Kunming mice, female, aged 8-10 weeks, weighing approximately 26g, were kept on standard laboratory conditions.Tumor cell line; human gastric cell line; SGC-7901.Agents: interferon-r Staphylococcin Aureus Injection, NDV-L, LPS, activated carbon suspension, RPMI 1640,TRIzol agent, Access RT PCR System , Taq DNA polymerases, Rsa I, PCR Cleanup Kit, QIA quick Gel Extraction Kit, pGEM -T easy vector system II, MTT agent,DMSO.Instrument: Surgical operation apparatus, digital camera, colored analysis system of pathologic diagram, cell house, ultraviolet - visible spectrophotome-try, PCR thermal cycler, horizontal electrophoresis, quantitative DNA amplic-tion/Detecting system, electromicroscope.METHODSThe experiment is divided into three partsAnimal group: The mice were divided into experiment group and control group. The experiment group was devided into 3 subgroups. I (IFN-r): Each time 2 ml IFN-r (10000U) was administrated ip; G ( Staphylococcin Aureus Injection) ; Each time 2 ml Staphylococcin Aureus Injection (500U)was administrated ip; N (NDV-L); Each time 2 ml NDV-L (50HU) was administrated ip. The control group is P (PBS) : Each time 2 ml PBS was administrated ip.
