Surgery, chemotherapy and radiotherapy are still main therapies of carcinoma. Recently, with respect of studies on mechanisms of anti-carcinogens,approach for the suppression of carcinoma's neovascularization become an very important area of new anti-carcinogens' research and development. Previous studies demonstrated that these drugs not only have therapical capability, but also have prophylaxes for metastasis. It is an effective way for the development of new anticarcinogens by exploiting modern technical methods for the research of suppreesion of carcinoma's neovascularization activities ofChinese material medica.In this study, active component M12 was separated from Muhuitou (diversifolious Patrinia Root), a traditional Chinese medicine. We investigated its effects on endothelial cells ECV-304 of human umbilical vein. It was observed that M12 has strong suppression activities against cells' proliferation. The results of studies on its mechanisms implied that ECV-304's apoptosis facilitated by M12 may account for the suppression of cells' proliferation. Meanwhile, from gene expression spectra assay of human umbilical vein entothelial cell ECV-304 and human heptocarcinoma cell line BEL-7402 by gene chip technologies, differential expression genes, i.e. M12 activity-related genes, were screened. Its mechanisms were investigated on the basis of gene level. Assay of effects on angioges.is of chick embryo chorioallantoic membrance showed that M12 couldsuppress or destruct its neovascularization. It can be seen that M12 could suppress effectively growth of cancer from the investigation of M12 against H22 liver - in situ carcinoma. This result is dramastically different compared with the control group, probably due to M12-stimulated production of large amounts of INF-a and IFN- cytokines to kill tumor cells.
