| Objective:To study the related gene expression in primary hepatocellular carcinoma of different stages(cancerous tissue,paraneoplastic tissue,normal tissue)by cDNA microarray technique,Looking further at different stages of primary liver cancer tissues differentially expressed genes,its possible molecular mechanisms for analysis.Methods:(1)The clinical data of patients with primary hepatocellular carcinoma will be collected to establish a database and tumor tissue specimen bank,Screening strict standard of 20 cases of hepatocellular carcinoma,cancerous tissue and normal tissue.The expression profiles of 20 cases of primary hepatocellular carcinoma at different stages of sequentially compares;(2)Extraction of total RNA 20 samples of different stages,quality control agarose gel electrophoresis,fluorescently labeled purified total RNA,reverse transcribed into cDNA,using a single standard method Cy3-dUTP labeled in hepatocellular carcinoma tissues,adjacent tissues and after the probe is made of normal tissue,and after washing cDNA microarray hybridization chip,followed by scanning the chip fluorescent signals by SAS software for analysis of microarray image,and finally get differential gene screening.Results:(1)Through SAS software analysis:Cancerous tissue and cancerous tissue differentially expressed genes 1501,Cancer tissue and normal tissue differentially expressed genes 2059.(2)The screened differential expression genes in different stages of hepatocellular carcinoma were mainly involved three categories according to GO Category,namely,Catalytic activity,ion channel signal transduction,immune response,activity regulation,transcriptional activity regulation,and protein synthesis,decomposition,transport processes,cell growth,apoptosis and other functional processes;(3)We found that several major Pathways in different stages of esophageal carcinoma were involved in Mechanism of Acetaminophen Activity and Toxicity.Leukocyte transendothelial migration-(human)pathway,TSP-1 Induced Apoptosis in Micro vascular Endothelial Cell Pathway,cdc25 and chkl Regulatory Pathway in response to DNA damage,Transcriptional activation of dbpb from mRNA pathway.Conclusion:(1)3560 differential expression genes were found in different stages(cancerous tissue,paraneoplastic tissue,normal tissue of hepatic carcinoma by gene chip technology.(2)Differences in gene expression studies are expected to provide a new basis for the diagnosis and treatment of primary hepatic carcinoma targets;(3)With analyzing the signal pathway of differential expression genes in hepatic carcinoma,molecular signal transduction mechanism of them was preliminary revealed. |
