The mechanism whereby the kidney replenishing herb promote the secretion of prolactin and the effect of this herb on the NK cells in the model of bromocriptine-induced abortionPhD student: Lu FangxianAdvisor: Gui SuiqiSection one: The possible mechanism whereby the kidney-replenishing herb promote the secretion of prolactin Objectives: Kidney-replenishing herb has been applied to treat for recurrent spontaneous abortion which is due to the dysfunction of immuno-endocrine network at the maternal-fetal interface. Conception rate in these patients amounts to 90% and the levels of serum P, PRL, β-HCG are increasing at the same time. These data indicate that kidney-replenishing herb has a positive effect on patients with abortion through promoting the secretion of key hormones during pregnancy, for instance, prolactin. In order to elucidate the detailed mechanism of the kidney replenishing herbs, we established the improved animal model of bromocriptine-induced abortion, which can simulate the dysfunction of immuno-endocrine network. In the past studies, we have found that kidney -replenishing herb can lower the abortion rate through increasing the expression of prolactin-receptor, progesterone receptor and estrogen receptor in the deciduas and ovary, promoting the secretion of prolactin, progesterone during pregnancy, and balancing the Th1 type cytokines and Th2-type cytokines at the maternal-fetal interface. The objective of this study is to explore the mechanism whereby the herb can promote the secretion of prolactin, we observed the effect of kidney replenishing herb on the expression of PrRP and PrRP-R in the improved animal model of bromocriptine-induced abortion. Methods: Pregnant SD rats were divided into five groups in random, group A was injected with bromocriptine (0.3mg. kg-1.d-1 during the pregnant day of 6-8. Group B, C, D were taken with the powder of Chinese herb (3g.kg-1.d-1), PRL(8IU each time, twice a day)and progesterone(8mg. kg-1.d-1) respectively in the pregnant day of 1-11 except that these three groups were all injected with bromocriptine in the pregnant day of 6-8 like group A. The rats of group E were normal pregnant rats. All these rats were killed on the pregnant day of 12 to observe the pregnant rates of five groups. Meanwhile, RT-PCR was applied to analyze the mRNA transcription of hypothalamus, decidualand placental PrRP, and expression of pituitary, placental and decidual PrRP receptor, and the expression of pituitary prolactin receptor. Results: When the concentration of bromocriptine was 0.3mg.kg-1, the rate of abortion in group A was 75%, and the pregnant rate was 25%. The pregnant rate and the number of litters of group B were 100% and 103 respectively, which were significantly higher than those of group A(P <0.01). The results indicated that the powder of kidney-replenishing herb had a positive effect on the abortion. The pregnant rates of group C, D, E were 91.7% , 91.7% and 100% respectively, which were also significantly higher than that of group A. The number of litters in group C (153), D (148), E (158) were also significantly higher that that of group A (70). PrRP was determined only in the abortive deciduas of group A, and the PrRP expression on hypothalamus of group A was significantly lower than that of group B, C, D, E. PrRP receptor was all expressed in the ovary and placenta of five groups, but there were no significantly difference. The expression of PrRP-R, PRL-R on the pituitary of group A was significantly higher than that of B, C, D, E group(P<0.05). Conclusions: 1. Kidney replenishing herb has an obvious effect on SD rats of the bromocriptine-induced abortion. 2. During the pregnancy, hypothalamic tissue is the main site to compose PrRP and PrRP can hardly be secreted by placenta and ovary. 3. Owing to the expression of PrRP receptor in deciduas and pituitary, we can speculate that kidney replenishing herb can promote the pituitary and decidual prolactin through affecting the composition of PrRP from hypothalamic tis...
