ObjectivesTo explore the charateristics of microvascular architecture in human hippocampus to provide the basis for the prevention and treatment of hippocampal is-chemic diseases , and explore the characteristics of the hippocampal morphology to provide a theoretical basis for the surgical approach and vascular interventions of the hippocampal region. To examine whether Thioredoxin has a cure effect of antioxidative treatment on hippocampus of ischemic - reperfused mice.Methods1 . With arteries infused by a mixed liquid of ink and formaldehyde , all the specimens were cut into small slices and were examined by the image analyzer.2. With a common carotid artery infused by methacrylate resin and eroded by thick hydrochloric acid, following the distilled rinsing ,the charaterstics and diameters of human microvascular arteries were examed and measured with scanning electron microscope and micrometer, respectively.3.28 hippocampal regions coming from the formaldehyde -fixed autopsies were obtained. The microanatomy was performed under the Olympus surgical microscope (6×20);The origins and distributions of the arteries were also examined , including the adjoining areas. The diameters were measured with micrometer.4. In our present study, focal brain ischemic - reperfused mice model was used. 8 - week BALB/c mice were grouped randomly as : control group , shame operated group , PBS - treatment group , and Thioredoxin - treatment group. An-imals were subjected to focal brain ischemia by cervical artery occlusion 15 min, and killed at 24 hr after reperfusion. Western Blotting and RT - PCR were used to detect cellular protein oxidation and c - Fos mRNA expression, respectively.Results1. There were two microvessel layers in the hippocampal dentate gyrus , called the deep layer and the superficial layer. What's more, vascular neural columns were also within these regions. There were artery - artery and artery -vein anastomoses. The vascular densities of dentate gyrus and deep layer of CA1 were 0. 365 ± 0. 018μm2/μm2,0. 246 ± 0.017μm2/μm2, respectively. There were two vascular layers and artery - artery and artery - vein anastomoses in the hippocampus.2. The human hippocampal microvascular artery included as follows: perforating arteres, anterior microvascular arteries, middle microvascular arteries, and the posterior microvascular arteries. The outer diameters were 200 - 60 um , 80 -30um , 30 -20um and 20 -10um , respectively. The fixed specimens were examed under the scanning electron microscope and annular , bead -alike and caulm - alike impressions were observed.3. Arteries supplying the anterior part of the hippocampus were as follows: the perforating branches coming from the original part of the anterior cerebral and middle cerebral arteries and the perforating ones from the anterior choroidal artery. The average diameter of each artery was 0.65 0. 17mm, 0. 77 0. 10mm and 0. 43 0. 06mm, respectively. Three arterial groups supplying the head of hippocampus were defined: hippocampal branches originating from the anterior choroidal (diameter, 0.41 0. 14mm) , posterior cerebral (diameter, 0.43 0. 08mm) and anterior inferior temporal (diameter, 0.48 0.10mm) arteries. Arteries irrigating the body of the hippocampus were hippocampal arteries coming from posterior inferior temporal, posterior cerebral and lateral posterior choroidal arteries, respectively.4. At 24 hours after cerebral arter occlusion, infracted area and volume were significantly smaller in Thioredoxin - treated group than in PBS - treatedgroup mice. Protein carbonyl content, a marker of cellular protein oxidation, in Thioredoxin - treated group mice showed less increase than that of PBS - treated group mice. Moreover, the mRNA expression of c - fos was stronger in Thioredoxin - treated group mice than in PBS - treated group mice.Conclusions1.
