Experimental Study Of Acupuncture Preconditioning Effect On Protection Of Rat's Brain Ischemia

To explore new preventing and cure method of cerebro-vascular disease and preventive mechanism of acupuncture preconditioning, this experiment divided 152 rats into seven groups randomly: normal control group, sham-opration group , ischemic preconditioning control group, ischemic group , ischemic preconditioning group , acupuncture preconditioning control group , acupuncture preconditioning group. 12 , 24, 48 , 72h after operation, use HE staining to observe change of histomorphology, neuronal Nissl's staining to observe neuronal damage, TUNEL method to display apoptosis state, electron microscope to observe ultrastructure, immunohistochemistry and ISH technique to detect HSP70, Bcl-2, c-fosmRNA and protein expression,wet-dry comparison to detect moisture content , biochemical method to detect ADO of brain tissure. (1) Acupuncture preconditioning has no influence on histomorphology. (2) Compared with ischemic group, the neural damage and dropsy is light, apoptosis cells are few, recovery of neural form is rapid in cerebral cortex and hippocampal CA1 section of acupuncture preconditioning group,similar like ischemic preconditioning group. (3)Compared with normal control group, HSP70, Bcl-2, c-fosmRNA and protein expression are increased in cerebral cortex and hippocampal CA1 section of acupuncture preconditioning control group. (4)Compared with ischemic group, the appearance of HSP70, Bcl-2, c-fosmRNA and protein expression are earlier and duration are long in cerebral cortex and hippocampal CA1 section of acupuncture preconditioning group. (5)Compared with ischemic group, moisture content of brain tissure is decreased and endogenous adenosine is increasing of acupuncture preconditioning group.Conclusion: 1.Acupuncture preconditioning can induce BIT, lighten neural damage of ischemic cerebrovascular disease.2.Acupuncture preconditioning can prevent apoptosis in cerebral cortex and hippocampal CA1 section.3. Acupuncture preconditioning can prevent apoptosis after severe brain ischimia by increasing Bcl-2, c-fosmRNA and protein expression.4.Induce HSP70mRNA and protein expression is one of preventive mechanism of acupuncture preconditioning .
