It was found that acupuncture of neiguan points could ameliorate cardiac fuction on coronary heart disease patients in clinical practice ,and it is in experimental study from the aspect of amelioration of myocardial energy metabolism , diminution of oxygen-derived free radicals and adjustment of vessel endogenous protection subtance and so on. But there is no literature to generally, cellular and molecular level about acupuncture on the different meridians point for treating injury of myocardial ischemical reperfusion with the approach of regulating mechanisms of calcium overload.Objectives: In order to study regulative mechanism of calcium overload as a whole and to cellular and moleclar level. Try to have a comparative study between Pericardium , heart Meridian and large intestine Meridian; To explore specificity of the correlation between Hand-Jue Yin Pericardium Meridian and Heart , Meridians and viscera.Methods: All rats were randomly divided into five groups including pseudo-operation group, model group, electroacupuncture on Neiguan group, electroacupuncture on Shenmen group , electroacupuncture on Hegu group.Classical animal model of myocardial ischemical reperfusion injury in rats was adopted . Ekectrocar diogram elec-trocardiograph(ECG) was detectedfor observing changes of ST segment s T wave and odds of QX/QT;myocardial intracellular Ca was tested by spectrofluorometer with double wave-length; activity of Ca '-ATPase and Na+-lC-ATPase was examinated by method of fixing phosphorum; expression of Ca2t-ATPasemRNA and Na'-lC-ATPasemRNA were observed by method of reverse transcriptase Polymerase chain reaction(RT - PCR); changes of microstructure and ultrastructure of myocardial tissue was also observed under light microscopy and electron one.Results: (1)Heightening of ST segments T wave and odds of QX/QT were observed when rats animal model was built; EA on Neiguan and Shenmen acupoints could promote partly rehablitation of ST segments T wave and odds of QX/QT of ECG, especially on B, whereas there existed a weak effect on decrease odds of QX/QT on C and no obvious comparison with A(P>0.05). Furthermore, there existed a vague effect on ST segments T wave and odds of QX/QT and no distinct comparison with A(P>0.05).(2)Augment of myocardial intracellular Ca2+ were observed when rats animal model was built; EA on Neiguan point could inhibit calcium overload, that is strongly different from EA on Shenmen or Hegu point(P<0.01), however, such effect was weaker than that on E(P<0.05). There had a tendency for decreasing myocardial intracellular Ca2+ on C and D, while no clear difference from A(P>0.05).(3) Activty of calcium pump and sodium pump was reduced when rats animal model was built; EA on Neiguan point could inhibit such two enzymes' activity to some extent, that is strongly different from EA on Shenmen or Hegu point(P<0.01), EA on Shenmen point also had such effect on restraining activity of calcium pump, but it was weaker than B(P<0.01). EA on Shenmen and Hegu point had no clear effect on activity of sodium pump, while there existed no clear difference from A(P>0.05).(4)Calcium pump and sodium pump's gene expression was reduced whenrats animal model was built; EA on Neiguan point could, that is strongly different from A (P<0.01), EA on Shenmen point also had such effect on increasing Calcium pump's gene expression, but it was weaker than B(P<0.01). EA on Shenmen and Hegu point had no clear effect on activity of sodium pump's gene expression, while there existed no clear difference from A(P>0.05).(5)EA on Neiguan point could promote more efficiently rehabilitation of microstructure and ultrastructure of myocardial tissue with myocardial ischemical reperfusion injury, EA on Shenmen point also had such effect, while EA on Hegu point had no clear effect.Conclusion: (1) Myocardial cell cytoprotection and inhibiting calcium overload could Realize by EA on Neiguan point for increasing calcium pump and sodium pump's gene expression and activity, reducing content of myo...
