Objective: To study the effect of repairing full-thickness articular cartilage defects using osteoperisteal grafts and bone marrow mesenchymal stem cells(BMSCs) transfected with ad5/CMVTGF- 1bone.in rabbitsMethods: 1. The MSCs are isolated from the bone marrow, purified and cultured. And then the biological features are further studied; 2. Ad5/CMV-TGF 1are constructed; 3. BMSCs are transfected with Ad5/CMV-TGF then the expressing of TGF 1are evaluated;4. The full-thickness articular cartilage defects are repaired using osteoperisteal grafts and BMSCs transfected with ad5/CMV-TGF-The defects are divided into two groups (group A and control B)according to the BMSCs(gene modification group and no gene modification group). The repairing of the cartilage defect and the expression of the TGF1 are evaluated grossly, histologically and immunohistologically 4, 8, 12, 24 weeks after operation respectively.Results: 1. The proliferation and expanding of BMSCs are extremely active in the culture. During the culture the number of the MSCs were on increase. 2. Ad5/CMV-TGF1 are got in 293 cells with using plasmid of pACCMV TGF containing the TGF gene of cDNA and the plamid pJM17 by transfected, and titer high; 3. The expressing of TGF are about 8 weeks; 4. Gross observation: The repairing tissues Group A are excellent 12 weeks and maintain 24 weeks after operation. The repairing tissues of group B are inferior to that of group A and degenerative 24 weeks. Histology observation: New tissue of group A is of cartilage tissue which resembled the ordinary joint cartilage very much, and that of groupB is alike of cartilage tissue.Conclusion : The experimental study of repairing full-thickness articular cartilage defects using osteoperisteal grafts and bone marrow mesenchymal stem cells transfected with ad5/CMV-TGF1 is proved to be feasible. However, further studies such as improving biomaterial design, evaluatingthe biomechanical properties of the regenerated tissue and ensureing clinical safety et al. are necessary to enhanced the efficiency of the idea.
