DO(distraction steogenesis, induction of osteogenesis by means of an osteotomy followed by fixation with an external fixation and subsequent controlled distraction of the callus) is a useful technique with widesoread clinical application in the treatment of bone defects, limb deformities and fracture non-unions. The aim of distraction osteogenesis is to obtain optimal bone-lengthening. Despite the use of an appropriate distraction rate, formation of new bone is not always optimal and the long duration of the bone consolidation phase of distraction osteogenesis treatment can be a cause of considerable morbidity for the patients, and in clinical practice, there is a burning need for augmentation of bone consolidation during distraction osteogenesis. To facilitate maturation of the regenerated bone, several experimental approaches have been described, included mechanical stimulation, electrical stimulation, transplantation of bone-marrow cells or osteoblast-like cells, and stimulation with recombinant fibroblast growth factor and vitamin D3 . Bone formation in distraction osteogenesis resembles aspects of both normal fracture healing andembryonic limb developments. Although many cytokines, growth factors, hormones and extracellular matrix components are capable of regulating specific aspects of bone regeneration and remodeling during bone growth and repair, BMPs are among the most potent of the osteoinductive factors . Urist made the key discovery that demineralized bone induces new bone formation when implanted intramuscularly. Subsequent purification of bone morphogenetic proteins (BMPs) has opened a new venue for skeletal tissue engineering. BMPs includes BMP-1, BMP-2, BMP-3, BMP-4, BMP-5, BMP-6, BMP-7 and soon. Among the BMPs, BMP-2 has been shown to act on the proliferation and differentiation of osteoprogenitor cells into bone-forming cells. The therapeutic potential of recombinant human bone morphogenetic protein-2 (rhBMP-2) has been examined in animal models of critical-sized bone defects, osteochondral defects, and spinal fusion, but there is less report about the relation of BMP-2 and distraction osteogenesis. Therefore, we study the effect of rhBMP-2 and basic fitiroblast growth factor(bFGF)on the proliferation and differentiation in vitro culture of bone mesenchymal stem cells (BMSc) , test the ability of rhBMP-2 to accelerate the consolidation phase of distraction osteogenesis in a rabbit model of leg lengthening.1 The culture and the proliferation and differention in vitro of bone mesenchymal stem cellsobjective : to study the growth and the differentiation of BMSc. Methods: The BMSc were obtained 1ml aspirate from iliums of rabbits , then BMSc were cultivated in vitro. We observed thegrowth characteristics of BMSc under the microscope. We tested the survival and increase of BMSc growth by the relative data from MTT method,detected the rate of ALP(+) by the improved Gomori' s method. Results: The adherent BMSc gave rise to colonies and grew quickly. The appearances of BMSc were not altered after several passages. Some colonies and cells locating in the center of colonies expressed significant ALP(+) , then mineralized plagues were detected. Conclusion: The BMSc were obtained easily and The BMSc has high capability to proliferate and differentiate in vitro. 2 rhBMP-2,bFGF affect the proliferation and differentiation in vitro culture of bone mesenchvmal stem cells Objective:To investgate the effect of rhBMP-2 and bFGF on the differentiation and proliferation of cultured BMSc.
