Establishment of PSGL-1 Expressing cell line and studies on its morphology and biological functionsP-Selectin glycoprotein ligand 1(PSGL-1) is distributed on the surface of leukocytes, and binds to P-, L- and E-selectin in vitro. PSGL-1 mediates leukocyte tethering and rolling during inflammation by binding to P-selectin on the endothelial cell surface.We have established a melanoma A2058 cell line that stably expresses human PSGL-1. The results showed that PSGL-1 induces cell aggregation and morphological changes compared with vector-transfected A2058 cells, including a significant reduction in cell size and a dramatic alteration to membrane architecture. Molecular analysis showed that PSGL-1 not only up-regulated expression of N-cadherin and (-catenin, but also induced their redistribution in the cells to cell membrane. Adhesion assays demonstrated that PSGL-1 reduces cell attachment to tissue culture plates but increases cell attachment to culture plates pre-coated with versican G3 and fibronectin, two extra-cellular matrix molecules. Further studies showed that PSGL-1 reduced cell mobility during wounding assays. Taken together, our results suggest PSGL-1, a transmembrane protein, could regulate cell mobility, attachment and aggregation through different, novel mechanisms.
