PART ONE Autologous Skeleton Satellite Cells Transplantation to Treat Coronary Heart DiseaseFirst section Culture and identification of skeleton satellite cell Objective: To establish the methods for culture and identification of skeleton satellite cell. Method: Young porcine satellite cell were obtained by the methods of mechanic and enzymic digestion. Satellite cells were identified with cellular immunochemical stain and reverse transcription polymerrase chain reaction(RT-PCR). Result: satellite cells were spindle-shaped and adherea on the bottom of the flask after 48 hours culture.if fetus bovine serum concentration decreased to 2%, satellite cells commenced the fusion process, which resulted in formation of multinucleated myotubes. Skeleton satellite cell were postive with cellular immunochemical stain Desmin. They also expressed hepatocyte growth factor receptor c-met gene. Conclusion: This methods was suitable for collection skeleton satellite cell.
