Objective To investigate the effect of androgen with different concentration on gene and protein expression of desmin in satellite cell of skeletal muscle. Methods Primary skeletal muscle satellite cell and cultured by enzymatic digestion methods. The morphology of skeletal muscle satellite cell was studied by inverted phase contrast microscope. Its molecular markers were observed by desmin and a-actin immunocytochemistry. Primary3~4days cells were used in experiment, and they were randomly divided into group zero (control group).group one(0.01×10-3ng/ul androgen group),group two(0.04×10-3ng/ul androgen group), group three(0.16×10-3ng/ul androgen group),group four(0.64×10-3ng/ul androgen group),groupfive(1×10-3ng/ul androgen group). The change of morphology of skeletal muscle satellite cell was observed by inverted phase contrast microscope. Applicating semi-quantitative PCR and Western blotting to measure the expression of desmin gene and desmin by skeletal muscle satellite which are interfered by different concentrations of androgen. Results The espression of desmin gene and the espression of desmin protein were increased with the increasing of the concentration of androgens. Conclusion Skeletal muscle satellite cell are round and a few cells are irregular, which are similar with fibroblasts in morphology. The desmin mRNA/GAPDH mRNA:group0:1.093±0.003; groupl:1.470±0.003; group2:1.691±0.004; group3:2.216±0.005; group4:2.731±0.003; group5:3.205±0.004。The desmin/GAPDH of every group:group0:1.125±0.007; group1:1.374±0.005; group2:1.395±0.004; group3:1.446±0.005; group4:1.685±0.006; group5:1.835±0.006。The espression of desmin gene and the desmin could be promoted by androgen. By the increasing of androgen, the promotion of desmin was stronger.
