BACKGROUND The metastasis and recurrence is the important death reason of breast cancer patients, and is the difficult problem for clinical treatment. The metastasis of breast cancer is most complicative multiple> sequenced progress, and is the results of different factor modulation. With the using of gene chip, using gene chip in the research of breast cancer metastasis genes is the hot dot of recent breast cancer research. On the base of gene chip , cinical test for the metastasis relative gene is the most part of the metastasis research. OBJECTIVE To pursue the relativity between the expression of MMP-2 and FN with the metastasis of breast cancer.METHOD Using the different expression genes MMP-2 and FN, with flurence-quantitative RT-PCR and western blot methods, analysis the mRNA expression relations of MMP-2 and FN with clinical prognostic factors in 116 clinical samples and protein expression in 7 breast cancer cell lines.RESULT The mRNA expression of MMP-2 and FN is up-regulated in primary samples, down-regulated in metastatic samples. The mRNA expression of MMP-2 is unrelated with lymphnode metastasis, but the mRNA expression of FN is up-regulated in metastatic lymphnodes. The mRNA expression of MMP-2 and FN is up-grade one, higher than in grade two and three. The mRNA expression of MMP-2 is up-regulated in tumor=2cm, higher than in tumor > 2cm, tumor > 5cm;but the FN mRNA expression is irrelated with tumor size. The mRNA expression of MMP-2 is irrelated with clinical stage, but the mRNA expression of FN is up-regulated in stage three. The mRNA expression of MMP-2 is up-regulated in lymphnode 1-3 positive, mRNA expression ofFN is up-regulated in more than 8 lymphnodes popsitive. The mRNA expression of MMP-2 and FN is irrelated with pothology type. The mRNA expression of MMP-2 is irrelated with ER, PR, but the mRNA expression of FN is up-regulated in ER positive, PR positive. The mRNA expression of MMP-2 and FN is down-regulated in metastatic cell lines, but protein expression is up-regulated in metastatic cell lines. CONCLUSION The mRNA expression of MMP-2 and FN is down-regulated in metastatic lymphnodes, implicating that mRNA expression of MMP-2 and FN is negative feedback regulated. FN mRNA is up-regulated in positive lymphnodes. FN mRNA is up-regulated in more than 8 lymphnode positive, MMP-2 mRNA is up-regulated in 1-3 lymphnode positive, indicating that FN and MMP-2 mRNA expression is related with breast cancer metastasis. MMP-2 and FN mRNA is unincreased with grade increasing, indicating that MMP-2 and FN mRNA is unrelated with breast cancer invasion. MMP-2 mRNA is up-regulated in tumor=2cm, indicating that MMP-2 mRNA up-regulated in early stage of breast cancer. FN mRNA expression is up-regulated in ER, PR positive. MMP-2 and FN mRNA is down-regulated in metastatic cell lines and MMP-2 and FN protein is up-regulated in metastatic cell lines.
