Objective: To study the effect of ovarian cancer cells on expressions of Fn and HA of human peritoneal mesothelial cells (HPMC) .Methods: The conditioned medium(CM) of ovarian carcinoma cell SKOV3 was tested by enzyme linked immunosorbent assay (ELISA) for transforming growth factor--β1(TGF- β1). Under different conditions, Fn gene expression of HPMC was studied by reverse transcription polymerase chain reaction (RT-PCR), Fn protein and hyaluronic acid (HA) levels in HPMC cell lysates and supernatants were studied by ELISA.Results: TGF- β1 in SK0V3-CM was (236.37±21.82) pg/ml. Before incubation with SK0V3-CM (blank control group), Fn gene expression of HPMC was 1.33±0.14, Fn protein levels in HPMC cell lysates and CM were (173.69±16.16) ng/ml and (135.32±11.75) ng/ml, HAlevels in HPMC cell lysates and CM were (17.15±1.43) ug/ml and (7.36±0.82) ug/ml. Following incubation with SKOV3-CM, Fn gene expression of HPMC was 2.64±0.17, Fn levels in HPMC cell lysates and CM were (297.69±28.02) ng/ml and (279.58±26.69) ng/ml, HA levels in HPMC cell lysates and CM were (37.29±3.54) μg/ml and (18.57±1.93) μg/ml, which were all higher than those in blank control group (P<0.01). Following incubation of HPMC with SKOV3-CM plus TGF-β1 neutralizing antibody, Fn gene expression of HPMC was 1.90±0.12, Fn levels in HPMC cell lysates and CM were (231.64±22.18) ng/ml and (212.03±19.54) ng/ml, HA in HPMC cell lysates and CM were (25.53±2.49)μg/ml...
