Objectice:To explore colon motility and the effect of KangYanLing after multiple organ dysfunction syndrome induced by bacteria peritonitis in rats. Methods: Thirty Wistar rats were divided three groups, which were the control group, the MODS group and the treatment group. (1) The number of stool points in all groups were checked; (2)10-4mol/L acetylcholine(Ach) and 10-1mol/L potassium chloride were used as stimulus to estimate the amplitude change of circular muscle strips, the cell length of colon smooth muscle and [Ca2+]i of colon smooth muscule in the rats; (3) the changes of IP3 were estimated by Radio-immunity analysis methods in the three groups; (4) the changes of serum NO in all groups were observed ; (5) the morphologic changes of distal colon smooth muscle were observed by the electron microscope. Result: (1)The number of stool points in the MODS group was less than the control's (t=3.01,P<0.05) ; the number of stool in the treatment group was more than the MODS's(t=3.044, P<0.05); (2) amplitude change of muscle strips in the MODS group was lower than the control group's,and amplitude change of muscle strips in the treatment group was higher than the MODS group's. The cell length of smooth muscle in the control group, the MODS group and the treatment group were 75.1+19.8uM,113.2+30.4nM and 81.1+21.7uM respectively. After using 10-4mol/L acetylcholine(Ach) and 10-1mol/L potassium chloride respectively, the cell length of smooth muscle in the control group were 49.1+11.8uM and 69.2+15.7uM; the cell length of smooth muscle in the MODS group were 65.1+18.5uM and 87.2+23.7uM; the cell length of smooth muscle in the treatment group were 57.1+13.8jiM and 77.9+23.3^M. The data of the control group were significantly different from the MODS group's(P<0.05), and the data of the treatment group were greatly different from the MODS group's(P<0.05). (3) The concentration of [Ca2+]i and IP3 in the MODS group were less than the control's, but the concentration of NO in the MODS group were higher than the control's(P<0.05); the concentration of [Ca2+]i and IP3 in the treatment group were more than the MODS group's,but the concentration of NO in the treatment group were lower than MODS group'; (4)There were differences in the morphologic change of electron microphone in all groups. The cellular structures of colon smooth muscle in the control group were normal; the swelling and degenerated mitochondria, the ruptured bridges andlots of rough endoplasmic reticulum were found in the MODS group; the cellular structure of colon smooth muscle in the treatmentl group were greatly improved. Conclusion: Colon motility was weakened in MODS condition, which was related with increased NO and decreased IP3 . KangYanLing could accelerate the colon motility by increasing the concentration of [Ca2+]i and IP3 of smooth muscle cell and by decreasing NO ,which switched on simultaneously cAMP> cGMP signal system and inostiol lipid signal system.
