Bladder cancer is the most common tumors of genitourinary tract .The most important clinical chartarictar of it is higher incidence rate, higher recurrence rate. Many scientists have witnessed that the immunological function is lower in the patien with tumor and the activity of the tumor infiltrating lymphocytic cell is lower,the ability killing tumor cell decreased.Then the "Immunologic black hole theory" was bring up.It was widly accetped that the decrease immunological function in the local of bladder tumor could lead that immune escape.This may permit the tumor cell to survive and keep growth.It may be the main reason of the higher recurrence rate in bladder cancer.But the mechanisms of lymphocytic cell activity repression is unclear.Nuclear factor κappB (NF-κB)is an important transcription factor.It could specially bind on immunoglobulin light chain gene enhancer.It existed in many cell.By regulating the transcription of many kind of immunologic correlation code gene, such as cytokine ( interleukin-2), immunologic receptor (interleukin-2R).It played a very important role in regulating immunological function of body. By using cell co-cultureing system ,We studied the role of nuclear factor κappB in bladder tumor inhibiting T lymphocytic cell functional in vitro.This study was designed as two parts. First,co-cultureing T lymphocytic cell with EJ cell,T24 cell,irradiated EJ cell, irradiated T24 cell and urinary bladder transitional epithelium,and the proliferation idex index(PI) of T lymphocytic cell ,the percentage of T lymphocytic cell of NF-κB nuclear Positive staining, the level of IL-2, sIL-2R in the supernatant and membrane IL-2R of T lymphocytic cell,T lymphocytic cell apoptosis index were measured by using MTT assay,immunohistochemistry,enzyme-link immunosorbent assay and flow cytometer.The main was to understand the role of nuclear factor κappB in bladder cancer inhibiting T lymphocytic cell functional in vitro.Second,using NF-κB Regulator with different dosage to chang the NF-κB activety.we studied the correlation between NF-κB activety and IL-2 system in T lymphocytic cell,verificated the results of first part, and discussed the possibility of curing bladder neoplasms based on NF-κB system.The main results were as follows:1.Bladder cancer cell EJ and T24 could decrease T lymphocytic cell functional and increase T lymphocytic cell apoptosis in vitro.2. Bladder cancer cell EJ and T24 could decrease NF-κB activity of T lymphocytic cell.3. Bladder cancer cell EJ and T24 could decrease the level of IL-2 in the the supernatant and the expression of IL-2R of T lymphocytic cell4.By decreasing NF-κB activity,Bladder cancer cell EJ and T24 could decrease the expression of IL-2 and IL-2R,interference IL-2 signal transmission, decrease T lymphocytic cell functiona.5.By increasing NF-κB activity,the functional decrease and apoptosis increase of T lymphocytic cell caused by bladder cancer cell EJ and T24 could be reversed,in the same time,the apoptosis of bladder cancer cell EJ and T24 incresed.so, it is possible that curing bladder cancer based on NF-κB system.6. IL-2 signal system may be the important downstream pass of NF-κB system in the function regulation of immune cell.it could be the target of curing bladder cancer based on NF-κB system...
