| Object: To investigate the effect of lights of different wave-lengthes on themorphology, function and the change of growth factor secreated of retina pigmentedepithelium, then try to discuss the pathogeny of myopia.Method: We isolated fetal human RPE cells by enzyme digestion. To analyze thebiologic characteristic of RPE cells, we plotted growth curve and examine theultrastructure by transmission electron microscopy. We irradiated the RPE cells withlights of different wave-lengthes, and then assay cell livingness by MTT, examine theultrastructure by transmission electron microscopy. We assayed the concentration ofHGF,bFGF and TGF-β in the culture medium by enzyme-linked immunosorbentassay(ELISA). After extract the total RNA of RPE cells irradiated by chromaticlights for 24 hours, we detected the mRNA expression of HGF by RT-PCR. Inaddition, we add TGF-βto culture medium, assay cell livingness by MTT, detectedthe mRNA expression of HGF by RT-PCR. then irradiated these RPE cells affected byTGF-β with lights of different wave-lengthes , assay cell livingness by MTT,assayed the concentration of HGF,bFGF and TGF-β in the culture medium byenzyme-linked immunosorbent assay(ELISA).Results: Fetal RPE cells grow actively,and the ultrastructure of RPE cells is normalin vitro. Lights of different wave-lengthes has different effects on the growth of RPEcells. After irradiated by lights of different wavelengths, RPE cells have different celllivingness and morphology. The results of ELISA indicated that RPE cells can secretedifferent amount of HGF,bFGF and TGF-β at different time after exposed to lightsof different wave-lengthes. The mRNA expression map of HGF is different after theRPE cells irradiated by different lights for 24 hours. The different concertration ofTGF-βall can restrain RPE cell growth and restrain the mRNA expression of HGF.The effect of TGF-βon RPE cell is different after exposed to lights of differentwave-lengthes. TGF-βcan restrain the secretion of HGF and bFGF, but can increasethe secretion by itself. - 3-夿—¦å¤§å¦åšå£«å¦ä½è®ºæ–‡Conclusion: From our various studies, we conformed the biologic characteristic ofRPE cells and the reciprocity of growth factor secreated by RPE cells .
