Electroporation and G418 screening were used to to establish a 3T3-L1 cell line which can express mouse resistain stably. And obtained a Cell lines with mouse Resistin highly expressed; To design mouse Retn-specific siRNA and construct a group of plasmids for expressing various mouse Rein-specific siRNA in mammalian cells. Electroporation and G418 screening were used to establish 3T3-L1 cell lines that used for stable expression of mouse Retn-specific siRNAs.and obtained a cell line which are potent in mediating Retn-specific RNA interference in adipocytes.obesity mouse modles were establishment with high fat diets . The expression of retn gene mRNA and insulin signals were analysised in 3T3-L1 cell line, mouse fat tissue.Estimated the glucose uptake of them and understanded the effect of Retn gene on insulin resistance .
