Magnetic nanoparticle technology is the latest research in tumor earlier diagnosis and therapy. It has made great progress in cell model and became the effective method in tumor therapy after radiation therapy, chemical therapy and surgery. The technology includes magnetic nanoparticle material, platform of constructing drug carrier and assembly technology with target cell. Magnetic nanoparticle carrier with anti-tumor gene or drug would inhibit tumor cell proliferation and induce tumor cell apoptosis. In this study, we'll explore the more powerful special target material to cancer and the new preparation technology of magnetic nanoparticle to enhance the carrying amount and the stability while reducing the injury to normal tissue.in vitro and in vivo. Our investigation purpose:1. To research new target material for reducing the damage of organs and tissues, based on safety and effective.2. To study or combinate nanoparticle encapusled epirubicinee with new technique for carrier of dosage and stability in vitro and vivo.3. To assess the efficacy of EPI on hepatic maligmant tumor in clinical utility.4. Purpose of investigation was to provide inducing apoptosis in vitro and in vivo with local ablation of tumor for clinical application.1. PART A Study on basical investigation: two aspects1.1 Magnetic nanoparticle encaspulate epirubicineMaterial and methods: Poly (d,l lactic-co-glycolic acid) microspheres as biodegradablemicrocarriers,encapusled drug was made in General hospital of Guangzhou Commandand institution of nanoparticle in Zhongshan University. Combination physical andchemical charaterastics, and modification particle in size and diameter.Result: 1. The average diameter of nanoparticle is 136 4.2nm(n=1000), Poly (d,llactic-co-glycolic acid) microspheres as biodegradable microcarriers.2.Epirubicinenanoparticle was degradated in vitro .3. The releasing of epirubincin nanoparticle show that the drug can not effect the speed of releasing of nanoparticle in vitro.4. In-vivo trial for epirubicine nanoparticle can lengthen the time of physical blood. 5. The result of depression tumor trial had difference between the two groups. Conclusion: Fe3O4 powder is a small particle with diameter of about . It have superparamagnetism, strong magnetism response will induce when it is put in magnetic field, but it will no longer have any magnetism when out of magnetic field. Epirubicine magnetic nanoparticles have good electromagnetism absorbence, they can absorb electromagnetism energy and produce heat in electromagnetism field after location in body, which will increase temperature of those parts in body and kill tumor cells, but can take little effect to normal tissue.1.2 Basical research on magnetic nanoparticle encapusled EPI inducing to apoptosis in Hep6 cell line in vitro and vivo. Material and method1. Cell culture of Hep6 and implant tumor of animal was performed.2. NEPI was made in the department of pharmology, Qeneral hospital of Guangzhou Command and Zhongshan University.3. NEPI can induce Hep6 cell, VX2, C57 rat, and nude mice to apoptosis in vitro and vivo. Annexin V/PI was assessed for dermination quantity and quality of DNA, apoptosis ratio in Hep6 cell culture and HCC under the observation on ultrastrucre of apoptosis and transmission electroscopy and scanning scopy.Trail was divided into group 3 ,treatment, untreatment, and control. Results: 1. The apoptosis ratio in the experimental group was increased from 25% to 54c% after 24 hours. The apoptosis ratio in the experimental group, biaoroubixing group and control group was 78%, 53% and 2% espectively after 36 hours. There was significant difference(t=3.05, P<0.05) between the results of each group. The apoptosis rate and quantity of medicine presented positive relativity with time (r =0.96. P < 0.05). 2. Pathological and ultrastructre displayed the largest necrosls in central zone of experimental HCC by NEPI.
