It is well known that cerebral vascular disease is the leading cause of death and long-termdisability in the world. Approximately 80% of all strokes are ischemic. It is extremely importantto study ischemic stroke. It is discovered that astrocytes and neurons secret many neurotrophic factors (NTFs) thatregulate the survival, differentiation and maintenance of function in neurons after stroke. NTFscan antagonize ischemic brain injury and promote the survival and regeneration of neurons inmany ways. Whereas, NTFs are large molecules, and can not get through the blood-brain barrier(BBB).Therefore, it may be an important approach to treating ischemic neuron injury by fullyinducing the expression of endogenous NTFs and their receptors. Based on the knowledge above, we carry out the study on the neurotrophy mechanism ofqingkailing effective composition antagonizing focal cerebral ischemic injury in rats. The articleincludes two parts. In the first part, we review the study on the model of middle cerebral artery occlusion(MCAO), the relationship between cerebral ishemia and glial reactions, the expression ofneurotrophic factors and the mechanism of neuroprotection against ischemic injury, the currentstudy on Chinese herbs decoction. The MCAO model is widely used for the cerebral ischemia in rats. The key to perform themodel is to control the weight and age of the rats. Moreover, the family of the animal and themethod of dealing with Nylon can affect directly the success rate. Glials become active after ischemia. The reason of neuronal death maybe is thedysfunctions in glials. The activation of glials after ischemia is extremely complex, which ischaracteristic with special model of time and space and whose effects include two parts. On onehand, active glials secret nitric oxide, TNF alpha, and other factors that can contribute toneuronal death, on the other hand, glials also secrete several neurotrophic factors that areimportant for neuronal survival and regeneration after stroke. The up-regulating expression of NTFs and their receptors is triggered by the activation ofNMDA receptor and the inner flowing of Ca2+.NTFs can prevent ischemic injury in many ways,which are as follows: maintain the density of Ca2+, antagonize the damage of free radicals,antagonize apoptosis, strengthen the activity of PKC, and repair the damaged neurons.-4- 清开灵有效组分抗大鼠局灶性脑缺血损伤的神经营养机制研究 The research of Chinese herbs decoction is a heat point. The methods and ideas of thestudy on Chinese herbs decoction become various. In the second part, we investigate the neurotrophy mechanism of qingkailing effectivecomposition antagonizing ischemic injury, and comprehensively analyze the experimentalresults and come to the conclusions as follows: 1. Qingkailing effective composition can promote the activation of astrocyts during theearly period after ischemia. Glial fibrillary acidic protein(GFAP) and S-100βare the markers of astrocytes activation.Qingkailing effective composition can upregulate the expression of GFAP and S-100βafterischmia for12h. The activation of Ast during the early period after ischemia plays an importantrole in antagonizing ischemic injury. Qingkailing effective composition can promote the activation of astrocytes specifically.And Jasminoidin upregulate the expression of GFAP, but Ruby mica, cholalic acid,compatibility of composition can upregulate the expression of S-100β. The activation of astrocytse has its restriction. Qingkailing effective composition canpromote the activation after ischemia for 12h, but the activation is inhibited after ischemia for24h. 2. Qingkailing effective composition can prevent ischemic injury through variousneurotrophy mechanisms during the early period after ischemia.
